Micro-fluidic Electrophoresis Chip (MFE Chip) is a rapidly developing field in recent years. It is a multidisciplinary technology and derived from the Micro Electro Mechanical System (MEMS), biochemistry and analytical chemistry. The micro-channel network on the chip is manufactured by the MEMS technology on the substrate of glass, fused silica, PDMS etc. Sample separation is performed in the micro-channel and detected by Optical or Chemical methods. It has been applied for life science, drug discovery and clinic medicine.In this dissertation, both electrophoresis principle on MFE chip and the design and development of MFE chip detection system are studied. A set of UV absorption detection system of MFE chip are also constructed. The experiments results showed that protein samples have been successfully separated and detected by using this system. The topics of this dissertation are presented below.1.The most recent developments of MFE chip system are reviewed and a novel design is presented;2. The design of MFE chip is laid out, a complete system are manufactured;3. The whole system is integrated by seven modules and optimized using a UV absorption based detection system;4. This detection system has been used for some protein samples and urine separation experiments, the results are well repeated and showed that it has met the need for protein rapid separation and detection.The dissertation showed that MFE chip has some advantages such as high speed and low sample consuming, as well as chip's design flexibility. But how to decrease the cost of MFE chip system and enhance it's detection sensitivity need further works.
