Image flow cytometry is a new concept which has been proposed by foreign scientists and technicians in recent years. It is an effective and innovative method to monitor algae by using image flow cytometry. Algae are one of the most important energy conversions in the marine ecosystem. They can go all over the world with ballast water, and it is closely related to the phenomenon of red tide, so it is important to monitor algae.In this paper, a method which combines the micro-fluidic chip and the smart phone to detect algae in ballast water is presented. The paper mainly implements the design of micro algae detection system based on image micro-fluidic. It has the merits of small size(8×4×5 cm), light weight (about 100g), and it can measure the single cell. It combines single lens with mobile phone, it has about 350 times magnification, resolution is about 1.5μm and FOV is about 150μm×150μm, it meets the sense of statistics and the requirement of detecting single cell at the same time. The main research content of this paper is:the design of single len optical microscopy, the design of digital image acquisition system, the design of real time video processing system, which includes optical module, smart phone based digital image acquisition module, image processing module, impedance pulse detection module; morphological characteristics of algae the cells were extracted, identified, and combined with the impedance detection technology to count algal cells through the microfluidic chip, we also complete the design of mechanical structure module, the modules are integrated together to complete the design of image flow cytometry detection device; And a simple code migration from the image processing program on PC to Android mobile phone is completed; in the end, the accuracy of the detection system is verified.
