Nature magazine awarded a new "technology of this century".It is called "lab on a chip".Microfluidic chip is one of them.The article in Nature magazine expounds the application of micro-nanofluidic chip from many aspects.Prospects and development process.Although the development of microfluidic chips is only a few decades,its application fields have been extended from simple sample analysis at the beginning to today's clinical diagnostic nucleic acid analysis,protein analysis,cell screening,etc.Microfluidic chip refers to the use of micromachining technology to produce microdevices integrated with microchannels,microvalves,micropumps,micromixing chambers,and microreaction chambers on a chip of a few square centimeters in size.Based on analytical chemistry,the microfluidic chip is loaded with reagents and biological samples to achieve sample injection,mixing,separation,reaction and detection.It is especially suitable for single molecule detection,separation analysis,rapid DNA sequencing and Research on protein enrichment.However,for microfluidic chips,how to align,combine and bond,and how to make them become key issues.Aiming at the problems of the existing micro-assembly system,this article introduces an alignment system for micro-nano fluidic chips,and independently designed a set of micro-nano fluidic chip alignment devices,and adopted a scale It is a chip assembly method of "sucking-clamping-rotating".The alignment and assembly application experiment was carried out based on the Hough line detection principle.Fabricated micro-nano fluid control chips,and conducted chip channel mixing experiments,and analyzed the experimental results.First,the current research status and existing problems of micro-assembly systems at home and abroad are analyzed.At the same time,it puts forward a plan of its own for specific chip processing and manufacturing,with the purpose of solving the problems of low chip bonding efficiency,slow manufacturing process,complex process,and difficult headlights.Secondly,in the process of micro-nanofluidic chip alignment,alignment accuracy,alignment method,alignment time,alignment theory,and whether the alignment process is convenient and efficient are issues we need to care about.Based on the above problems,using the Hough transform theory,this paper studies a micro-nano channel alignment method to complete the automatic alignment of the chip.Compared with some traditional chip alignment methods,it avoids the error and time-consuming process of the manual three-dimensional mobile platform in the chip alignment process.In addition,the Hough transform linear detection is applied to the alignment process of the microchannel chip and the nanochannel chip.The purpose is to achieve high-efficiency and accurate automatic alignment of micro-nano chips,and contribute to improving chip production efficiency and detection chip accuracy.Thirdly,based on the previous method of fabricating micro-nano structures on the same chip,although materials are saved,the accuracy of photolithography overprinting is high,and multi-configuration chips cannot be obtained.Now a new type of chip is built.The micro-nano fluid control chip automatic alignment device,which manufactures the micro-nano structure on two chips separately,so that combining the two chips at different positions not only can obtain more configuration micro-nano fluidic chips,but also reduces the requirements of manufacturing equipment and the complexity of the process are conducive to the realization of low-cost and batch micro-nano fluidic chips.The device is mainly composed of three parts: a clamping and adsorption part,a support part and a base part.The clamping force,adsorption force and other components that play an important role are calculated and checked.Design a practically applicable automatic alignment system and complete the assembly of the chip.In addition,in the process of aligning the glass chip with the micro-nano fluidic chip automatic manipulation system designed in this paper,the control accuracy,clamping force,and multi-degree-of-freedom displacement are several important factors that affect the quality of chip alignment.Therefore,these three High-precision and precise control of all aspects is one of the key problems solved in this paper.This chapter is based on the required design of the control part of the micro-nano fluid control chip automatic control system.STC15W4K32S4 is selected as the controller to control the pressure module and the motor module.The corresponding sensors are selected and the circuits of the control system are designed.Finally,part of the performance of the micro-nanofluidic chip alignment system was tested and the flow experiment of the chip was carried out.It mainly includes the calibration of the sensor of the pressure acquisition module,the straightness detection of the clamping and adsorption module,the production of the chip and the flow experiment.It proves the feasibility and reliability of aligning and fabricating the chip with the research method in this paper,and contributes to the research of micro-nano fluidic chip.
