Objective:In the foundation of chronic salpingitis model with qi stagnation and blood stasis,the research of Gexiazhuyu decoction treatment effect on chronic salpingitis model with qi stagnation and blood stasis,elaborates the mechanism of Gexiazhuyu decoction effect of TLR2,TLR4 and NF-?B signaling pathway,and explore its targets.Methods:The model of chronic salpingitis in rats with qi stagnation and blood stasis was established.The rats were randomly divided into blank group,operation group,model group,compound model group,patent medicine group,low,middle and high dose group of Gexiazhuyu decoction.On the first day,the model of chronic salpingitis was established with Staphylococcus aureus.On the 8th day,the model of qi stagnation and blood stasis was established.On the 29th day,the rats were treated with 28 consecutive days.General behavioral characterization,symptom score,histopathology,hemorrheology,and cytokine changes were observed.The mechanism of Gexiazhuyu decoction in the treatment of chronic salpingitis with qi stagnation and blood stasis was studied by molecular biology method.Results:The chronic salpingitis model with qi stagnation and blood stasis in rats was successfully constructed,after Gexiazhuyu decoction treatment,the chronic salpingitis rats with qi stagnation and blood stasis syndrome tended to be normal in their symptom evaluation and general behavior.The histopathological observation showed that the outer vessels of the fallopian tube were dilated,hyperemia and inflammatory cell infiltration decreased,and intraluminal exudate were also improved.Hemorrheology showed that compared with the compound model group,Gexiazhuyu decoction group,plasma viscosity,whole blood viscosity,reduced viscosity,erythrocyte rigidity index,erythrocyte aggregation index,erythrocyte sedimentation equation K value,erythrocyte electrophoresis time decreased,red blood cell deformability index increased.In addition,the cytokine levels were significantly improved.Compared with the compound model group,the ICAM-1,NF-?B,TLR2,TLR4,TNF-?,TNF-?,TGF-?,IFN-?,IL-6 of the Gexiazhuyu decoction group decreased significantly,and the IL-2 increased.Conclusions:1.Chronic salpingitis rats with qi stagnation and blood stasis can be induced by the method of salpinx injection of Staphylococcus aureus,harmonic,light,electricity,tail clip and slope.The symptoms are obvious and characteristic.2.Gexiazhuyu decoction can improve the body weight,appetite,general behavior,symptom score and pathological changes of fallopian tubes in chronic salpingitis rats with qi stagnation and blood stasis.3.Gexiazhuyu decoction can reduce the indices of chronic salpingitis rats with qi stagnation and blood stasis,including plasma viscosity,whole blood viscosity 3.0,30,50,100,180,reduced viscosity 3.0,30,50,100,180,erythrocyte rigidity index,erythrocyte aggregation index,erythrocyte sedimentation equation K value,erythrocyte electrophoresis time,can increase the red blood cell deformation index.4.Gexiazhuyu decoction can decrease the related signaling pathway factors TLR2,TLR4,NF-?B in rats with qi stagnation and blood stasis of chronic salpingitis.5.Gexiazhuyu decoction can reduce the inflammatory factors ICAM-1,TGF-?,TNF-?,TNF-? in rats with qi stagnation and blood stasis of chronic salp:ingitis.6.Gexiazhuyu decoction can regulate the immune factors of rats with qi stagnation and blood stasis of chronic salpingitis,increase IL-2,reduce IL-6 and IFN-?. |
