Objective:Studying the inflammatory index of IL-1 beta,PGE-2 and MMP-7 through the compound Bushen Lishi traditional Chinese medicine,we can describe its mechanism of action and providing a theoretical basis for the prevention and treatment of Kidney-tonifying and water-draining method of gouty arthritis.Methods:Healthy male SD rats were randomly divided into 7 groups after feeding for 7 days(n=84):(A)blank group,(B)model group,(C)Tongfengshu group,(D)colchicine group,(E)compound low concentration group,(F)compound middle concentration group,(G)compound high concentration group,which one group has 12 rats.The body mass per 100g rats 1 mL standard gavage,A and B given saline,C given Tongfengshu tablets suspension,D given colchicine suspension,E,F,G were treated with compound Chinese medicine low,middle,high concentration suspension orally,1 time a day,for 7 days.We use the uric acid sodium and potassium oxonate induced gouty arthritis rat model.After the infraorbital blood sampling of each group rats,serum separation,extract supernatant and serum from the sample extractions.Later,rats were killed after the double knee joint synovial tissue,making the slice,HE staining to observe the pathological changes.Elisa method was used to detect synovial tissues,which were detected in rat serum beta IL-1,ELISA PGE-2,MMP-7 content changes.Result:The HE pathological findings can be seen that,the blank group of synovial cells are in shape,arranged in neat rows.The model group shows urate crystals and the synovial cell morphology is not complete which with exudation.Compared with the control group,the level of uric acid in rats of model group are significantly increased(P<0.01)and the synovial cells are appeared MSU crystallization,which suggests that the rats model is successful.Compared with the blank group,the model group IL-1 beta,PGE-2,MMP-7 are significantly increased(P<0.01),with statistical significance.Compared with the model group,the treatment group IL-1 beta,PGE-2,MMP-7 are dropped significantly different(P<0.01).Compared with the compound middle concentration group of traditional Chinese medicine,the Tongfengshu group,compound low concentration group,compound high concentration group IL-1 beta,PGE-2,MMP-7 with a significant difference in the content(P<0.01),moreover the colchicine group IL-1 beta and MMP-7 with a significant difference in the content(P<0.01),while the content of PGE-2 has no differences(P>0.05).Comprehensive judgment,the Chinese medicine compound middle concentration group can significantly reduce the IL-1 beta,MMP-7 content with the best effect.As for the PGE-2 which the content decreased the most,the best curative effect is the compound high concentration group of Chinese medicine,with a dose-dependent trend.Conclusion:Bushen Lishi method Chinese herbal compound can inhibit the expression of IL-1,PGE-2,MMP-7 in rats with acute gouty arthritis,while the mechanism may be to inhibit the chemotaxis and activation of inflammatory cells,block the signal transduction pathway,inhibit the synthesis and release of PGE-2,MMP-7,IL-1 beta,which ultimately inhibit the acute inflammation of the joints.Therefore,it proves that the method of kidney-tonifying and water-draining method has a good effect on prevention and treatment of gouty arthritis,which also provides a new perspective for the prevention and treatment of the gout arthritis.
