| In this study,C57BL/6J mice were used as animal models to explore the effects of compound Kuding tea on growth performance,feed intake,liver and kidney function,blood glucose,lipid and molecular mechanism,and comprehensively evaluate the biological function of compound Kuding tea Thirty-six male C57/BL6 J mice with similar body weight were selected as blank control group(CN)high-fat diet model group(HFD)combined with Kuding tea 6mg/ m L(KL,normal tea drinking).Compound kuding tea 12mg/ml(KH,strong tea drinking)four treatment groups,each treatment 3 replicates,each replicate 3mice,a total of 18 w experimental cycle results are as follows:(1)Compared with the high-fat control group(HFD),the body weight of the low-dose compound Kuding tea group(KL,normal tea drinking)and the high-dose compound kuding tea group(KH,strong tea drinking)decreased by 11.4% and 16.0%,respectively(P< 0.05)and extremely significant(P<0.01);KL group and KH group had significant fasting blood glucose(P<0.05)and extremely significant(P<0.01)decreased and significantly improved impaired glucose tolerance.(2)In the experimental group combined with Kuding tea,the hyperlipid deposition induced by high fat diet was significantly reduced,and the compound kuding tea(KL,KH)significantly reduced the accumulation of TC and TG in liver tissue.(3)Compound Kuding tea can significantly improve the morphology of adipocytes and resist the whiteness of brown fat induced by high fat diet.Drinking compound Kuding tea with high fat diet can significantly reduce lipid deposition in liver cells.(4)The expression of UCP1,PGC-1 α,PPARγ,PRDM16 and CTP1 in KL and KH groups was significantly up-regulated compared with that in HFD group;Compared with the CN group,the expression of ATGL gene related to liver lipidysis was significantly decreased in the high-fat diet,and compound kuding tea could significantly improve the expression of ATGL gene in liver tissue,thus improving the ability of liver lipidysis Compared with CN group,the expression of FAS and ACC2 in SREBP-1C in KL and KH groups was significantly lower than that in HFD group.(5)For liver and kidney function of mice in each group,compound kuding tea significantly improved ALT and ALB levels in serum biochemical indices.At the gene level,compound Kuding tea significantly reduced the expression of apoptotic genes such as Casepase3 in liver,and induced the high expression of anti-apoptotic gene Bcl-2.In the detection of related indexes in kidney,compound Kuding tea first improved the serum biochemical CREA and UREA levels.The root cause may be that compound kuding tea inhibits the expression of apoptotic gene Bax in kidney,and induces the high expression of anti-apoptotic genes Bcl-2 and Bcl-xl.In conclusion,this study proves that long-term consumption of compound Kuding tea can effectively avoid health problems such as fat,hyperglycemia,hyperlipemia and obese non-alcoholic fatty liver induced by high fat diet,and is beneficial to liver and kidney functions to a certain extent.In the effect of reducing fat and losing weight,on the one hand,compound Kuding tea induces the expression of fat Browning gene,which promotes the Browning process of white fat,increases energy consumption of the body and thus reduces body fat consumption;On the other hand,by inducing the expression of liposolysis genes in the liver,inhibiting the adipose synthesis genes,regulating the glucose and lipid metabolism level of the body,achieving the effect of lowering lipid and glucose and reducing lipid deposition. |
