Obesity(Obesity)is often accompanied by the symptoms of hyperglycemia and hyperlipidemia,which is closely related to the pathogenesis of insulin resistance,non-alcoholic fatty liver and other diseases.Obesity seriously endangers animal health in animal production,mainly in endangering the perinatal health of cows and affecting the health of dogs and cats themselves.With the change of people’s eating habits and the improvement of living standard,the number of diseases such as hyperglycemia,hyperlipidemia and non-alcoholic fatty liver disease increases,and the risk of obesity increases year by year,which will further promote the occurrence of cardiovascular and cerebrovascular events.At present,there is no special effect therapy in clinic.Algae(Seaweed,S)has the effect of reducing fat and losing weight,but the therapeutic mechanism of seaweed is not clear.to investigate the therapeutic mechanism of seaweed on obesity,this study established a rat model of obesity by feeding a high-fat diet and treated it with aqueous solution of seaweed and alginate.to investigate the regulatory effect of seaweed on high-fat diet-induced obesity and related metabolic disorders in rats by detecting serum glucose metabolism,serum lipid metabolism,inflammatory pathways in liver and oxidative stress.In this study,the obese rat model induced by high fat diet,36 SD rats were random Ly divided into 6 groups,Six in each group,blank control group(NC),model group(HF),alginate group(65 mg/kg),positive control group(60 mg/kg),algal low dose group(5 g/kg)and algal high dose group(10 g/kg),Feeding 2 W,with high fat feed After successful modeling,4 consecutive W of administration,All rats were killed after 4 W.The HE staining was used to detect the type and degree of liver lesions,the serum lipid level and liver enzyme content were detected by biochemical method,and the expression of glucose metabolism level and TNF-α、IL-1β and IL-6 was detected by enzyme-linked immunoassay.The results are as follows:The effect of each drug on the weight and liver weight of obese rats: after 4 w of high fat diet,the body weight of the model group was significantly higher than that of the blank group(P < 0.05),and there was no significant difference between the body weight and the blank group(P > 0.05).Dog weight was significantly higher than in the blank group(P < 0.05),no significant difference between the dog weight and the blank group(P < 0.05).There was no significant difference in serum HDL level and blank group(P > 0.05)in each group and blank group(P < 0.05),low-density lipoprotein level in each group and blank group was significantly lower than that in model group(P > 0.05).The serum total cholesterol level in the control group and seaweed group was significantly higher than that in the model group(P < 0.05),there was no significant difference in serum total cholesterol in each treatment group(P > 0.05),and the serum triglyceride level in the model group was significantly higher than that in the blank group(P < 0.05).There was no significant difference between LDL、HDL、TG and TC levels and in the blank group(P > 0.05),LDL、TG and TC in the blank and treatment group,and significantly lower in the model group(P < 0.05).Serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)were detected: the ALT level of each treatment group and blank group was significantly lower than that of model group(P < 0.05),there was no significant difference between ALT level and blank group(P > 0.05),the AST of blank group and treatment group was significantly lower than that of model group(P < 0.05),there was no significant difference between rat AST level and blank group in each treatment group(P < 0.05).The liver weight of each drug group and blank group was significantly lower than that of model group(P < 0.05)after treatment with orlistat,sodium alginate and seaweed(P <0.05),the liver index of model group was significantly higher than that of blank group(P< 0.05),the liver index of each drug group was not significant(P < 0.05).The blood glucose level was significantly lower in each treatment group and blank group than in model group(P < 0.05),no significant difference in blood glucose level and blank group(P > 0.05),insulin level in each treatment group and blank group was significantly lower than in model group(P < 0.05),no significant difference in insulin level between treatment group and blank group(P > 0.05),glycosylated hemoglobin level in each treatment group and blank group was significantly lower than that in model group(P < 0.05)There was no significant difference between glycosylated hemoglobin level and blank group(P > 0.05),HOMA-IR level of treatment group and blank group was significantly lower than that of model group(P < 0.05),and there was no significant difference between HOMA-IR level and blank group(P > 0.05).The serum IL-1β levels in blank group and treatment group were significantly lower than those in model group(P < 0.05),serum IL-6 levels in blank group and treatment group were significantly lower than those in model group(P < 0.05),serum TNF-α levels in blank group and treatment group were significantly lower than those in model group.To sum up,the therapeutic effect of seaweed on obesity is mainly through the following three aspects: seaweed can reduce blood sugar,insulin and glycosylated hemoglobin levels in serum to improve insulin resistance in obese rats;seaweed can improve liver steatosis and reduce liver injury in obese rats;seaweed can inhibit chronic inflammation caused by obesity by reducing IL-1β、IL-6 and TNF-α.
