Zinc is the life element of animals and plays an indispensable role in the growth and development of animals.A series of diseases will appear in animal zinc deficiency,which will seriously affect carcass quality and animal husbandry development.It has been found that zinc deficiency is closely related to the pathogenesis of liver disease,and the liver is an important organ regulating zinc homeostasis in vivo.In recent years,fatty liver occurs from time to time in animals with a high incidence,and mainly occurs in animals with good nutritional status.Animal fatty liver can cause the decrease of egg production and feed utilization,and even cause death.Animal fatty liver has become a difficult problem for the development of animal husbandry.It is necessary to understand the pathogenesis of fatty liver and develop prevention and treatment methods.In this study,we explored the regulatory effect of zinc on hepatic glucose/lipid metabolism disorder in vivo and in vitro.In vitro experiment: zinc was added to hepatocyte culture medium;in vivo experiment: 8 week male C57BL/6 mice were divided into 4 groups(n=8): normal diet and normal zinc group(CON-30),normal diet and high zinc group(CON-90),high-fat diet and normal zinc group(HFD-30),high-fat diet and high zinc group(HFD-90).The regulation effect of high zinc on the disorder of glucose/lipid metabolism induced by high fat was studied by detecting the gene expression level,protein expression level and key enzyme activity related to glucose/lipid metabolism with molecular biological technology.In vitro studies have shown that zinc could significantly reduce triglyceride and total cholesterol levels and reduce lipid deposition in hepatocytes by regulating the expression of genes related to lipid metabolism and the activity of related enzymes.High zinc can alleviate the disorders of glucose/lipid metabolism caused by high-fat conditions,and has the effects of reducing glucose production,promoting glucose absorption and regulating energy metabolism.The results of in vivo study showed that high zinc could resist the increased lipid deposition caused by high-fat diet,inhibit the absorption of exogenous lipids,reduce the liver triglyceride(P=0.002)and total cholesterol levels(P=0.001)in the high-fat group,and significantly reduce the serum aspartate aminotransferase content(P=0.015)and serum total bile acid content(P=0.029)in the high-fat group to alleviate liver injury.From the experimental results,it is concluded that a high level of zinc can resist the increase of lipid accumulation induced by high-fat condition,reduce the lipid deposition in liver,and resist the disorder of glucose/lipid metabolism in liver,which provides a new perspective for the prevention and treatment of fatty liver in animals.
