| Purpose:To analyze the correlation between CKM,MYOM2 SNPs and the effect of resistance training.Picking out the sensitive molecular markers for resistance training.Using molecular markers to provide personalized resistance training programs for people with different physical conditions,and predicting the health benefits of individuals who are going to participate in resistance training.Method:Recruiting 242 Han college students,half males and females.Conducting resistance training to these 242 college students.The first round is a 12-week 5 X 10 resistance training program which include 5 sets of squat and bench press training with the 70%1RM intensity,and 10 times in each set.At the meantime,the total rest time between each two sets is 2 minutes.Warm-up and working takes about 40 minutes.The frequency of the entire process is 2 times a week for 12 weeks.Before and after the first round of training,the research will gain subjects’ body composition,muscle mass,and muscle strength,meanwhile drawing subjects’ blood.(Body composition indicators include body weight,lean mass,muscle,fat,bone mineral salt;other muscle mass indicators include upper limbs muscle mass,lower limbs muscle mass,trunk muscle mass,pectoralis major thickness,rectus femoris thickness;muscle strength indicators include squat maximum strength,bench press maximum strength,the peak extension force,the maximum extension work,the total extension work,the peak isometric contraction,the vertical jump height,the relative maximum power,the vertical jumping force divided by body weight).After the first round of experiment and data processed,we recalled the subjects with r<0.2(Cohen’ s Effect Size)in muscle mass or muscle strength.And the training program was replaced with the second round of 12-week 5 × 5 resistance training program.The training program only adjusted the number of repetitions and intensity of each group with 5 times and 85%1RM.The difference with the first round is that the research add one more test at the end of week eight.While conducting the first and second rounds of resistance training,white blood cell DNA was extracted,and the SNPs of the CKM gene and MYOM2 gene of the experimental subjects were detected by gene chip technology.After screening,the polymorphism information rs344793,rs1133190 of the CKM gene and rs12678739,rs2294066,rs3817705 of the MYOM2 gene were obtained.According to genotype grouping,normal test,single-sample T-test,non-parametric test are performed on body composition index changes,muscle strength index changes and muscle mass index changes.The initial values,change-values,growth rates of body composition,muscle strength,muscle mass are performed between groups analysis of variance,ANOVA analysis,non-parametric testing.Results:(1)After two round of resistance training intervention and result analysis,the research did not find polymorphisms at rs344793 and rs1133190 in CKM genes in initial values,change-values,growth rates of various indicators have significant difference about body composition and muscle strength.(2)The rs12678739 of MYOM2 gene was correlated with the growth rate of muscle mass.The growth rates of body muscle mass in the two rounds was separately,7.14%±7.02%and 9.51%±0.06%in the CC genotype group,5.50%±6.12%and 4.99%±4.08%in the TC genotype group,3.35%±5.52%and 3.47%±2.40%in the TT genotype group.The growth rates of upper limb muscle mass in the two rounds was separately,10.33%±10.21%and 12.87%±3.26%in the CC group,8.14%±10.06%and 3.65%±4.90%in the TC group,3.88%±9.82%and 3.84%±3.73%in the TT group.The first round of lower limb muscle growth rate was 9.19%±6.72%in the CC group,5.75%±6.40%in the TC group,3.69%±7.19%in the TT group;the second round of trunk muscle growth rate was 11.86%±3.36%in the CC group,and 5.59%±3.76%in the TC group,2.46%±3.96%in the TT group.The two rounds of the left Rectus femoris thickness growth rate were 16.29%±18.19%and 13.25%±9.41%in the CC group,9.86%±13.06%and 7.02%±9.00%in the TC group,and 4.67%±9.46%and 4.09%±8.79%in the TT group.The first round of the right Rectus femoris growth rate was 23.13%±22.59%in the CC group,13.19%±14.84%in the TC group,and 10.58%±12.14%in the TT group.(3)No correlation between the MYOM2 gene rs 12678739 polymorphism and the initial value,change and growth rate of various indicators of muscle strength;no correlation between the MYOM2 gene rs2294066,rs3817705 polymorphism and body composition,muscle mass,muscle the initial value,amount of change,and the growth rate of each indicator of power.Conclusions:(1)The research found that the MYOM2 gene rs12678739 polymorphism is correlated with muscle growth rate,which maybe used to guide people with muscle growth needs to choose resistance training programs(the muscle growth rate of CC genotype>the muscle growth rate of TC genotype>the muscle growth rate of TT genotype).According to results,the people of TC genotype choose 5 X 10 resistance training program and the people of TT genotype choose 5 X 5 resistance training program maybe gain more muscle growth.(2)The polymorphisms at rs344793 and rs1133190 of CKM gene and rs2294066 and rs3817705 of MOMY2 gene were no correlation with the initial value,growth quality and growth rate of muscle mass and muscle strength in the effect of resistance training. |
