Objective: To establish an acute closed soft tissue injury model in SD rats through the "quantitative gravity strike method",take self-made ointment as the experimental drug,and select Erhuangxinshang analgesic that has similar prescriptions and is widely used in clinical practice and has significant curative effect.Ointment is a positive control drug to observe injury symptoms and histomorphological changes of injured limbs in rats,and detect nitric oxide(NO),inducible nitric oxide synthase(i NOS)and structural nitric oxide synthase(c NOS)content changes,compare the anti-inflammatory effects of the two drugs,explore the mechanism of action,and lay a foundation for the self-made ointment to further reduce swelling and allergies,so as to further improve the Zheng's traumatology prescription system and provide the clinical application of the drug Provide theoretical basis.Methods:The successfully modeled SD rats were divided into 3 large groups according to the random number table method,namely,the model group(after model building,Vaseline was applied externally),the self-made prescription ointment group,and the Erhuang Xinshang Zhitong ointment group.Two hours later,the rats in each group were treated with external application of corresponding drugs on the local tissue damage.Routinely observe the mental state of each group of rats every day.Take 6 rats from each group on 1,4,and 7 days,weigh,observe the symptoms of the affected limbs and record the soft tissue injury symptom score,and observe the morphology of the local injury under a light microscope.Change,detect NO content in injured tissue(nitrate reductase method),i NOS expression(immunohistochemistry method)and activity(colorimetric method),and c NOS activity(colorimetric method),and the experimental data obtained are statistically based on SPSS25.0 Software analysis.Results:(1)Body weight: The body weight of the three groups of rats showed a trend of first decrease and then increase over time.Compared with the first and fourth days,the rats' body weight increased significantly on the 7th day of application,which was statistically significant.(P<0.05);The weight difference of rats between the groups was not significant and not statistically significant.(2)Score of soft tissue injury syndrome: The score of injury syndrome of the three groups of rats gradually decreased over time.On the 4th and 7th day of application,the tissue damage score of the self-made prescription ointment group was significantly reduced compared with the model group,which was statistically significant(P<0.05);compared with the model group,the tissue damage score of the Erhuangxinshangzhitong ointment group was The injury score was significantly reduced,which was statistically significant(P<0.05);the tissue injury score of the self-made prescription ointment group was slightly lower than that of the Erhuangxinshangzhitong ointment group,and there was no statistical significance between the two groups.(3)Observation of histomorphology: The morphology of muscle fibers in the three groups of rats gradually improved over time.On the 4th day of application,the model group showed muscle fiber degeneration and necrosis,inflammatory cell infiltration,and tissue edema.The self-made prescription ointment group and Erhuangxinshangzhitong ointment performed similarly,and the tissue morphology was significantly improved compared with the model group;on the 7th day,In the model group,muscle fiber edema and a small amount of inflammatory cell infiltration were still seen.The self-made prescription ointment group and Erhuang Xinshang Zhitong ointment showed almost no obvious inflammatory cells,and the morphology of muscle tissue was close to normal.(4)The content of NO in the injured tissue: In the comparison between the groups,the self-made prescription ointment group and the Erhuang Xinshang Zhitong ointment group,the 7th day of application compared with the 4th day of application,the content of NO in the tissue decreased significantly.There is statistical significance(P<0.05).Comparison between groups,at each time point of application,the difference between the self-made prescription ointment group and the Erhuang Xinshang Zhitong ointment group was not statistically significant.Compared with the model group,the NO content in the tissues of the two groups was reduced,which was statistically significant.Significance(P<0.05).(5)The activity and expression of i NOS in injured tissues: 1)The activity of i NOS: intra-group comparison,self-made prescription ointment group,Erhuang Xinshang Zhitong ointment group,comparison between the 7th day and the 4th day of application,i NOS in the tissue The activity of the drug was significantly decreased,which was statistically significant(P<0.05);the comparison between the groups,at each time point of application,the difference between the self-made prescription ointment group and the Erhuangxinshangzhitong ointment group was not statistically significant.Compared with the model group,the activity of i NOS in the tissue was significantly decreased,which was statistically significant(P<0.05);2)The expression of i NOS: The expression of i NOS detected by immunohistochemical method was mainly distributed in the cytoplasm and intercellular substance.In the model group,the self-made prescription ointment group,and the Erhuang Xinshang Zhitong ointment group,the expression of brown-yellow granules in the cytoplasm and intercellular substance of rats was found.Among them,the expression of i NOS in the self-made prescription ointment group was lower than that on the 7th day of application.4 days,the comparison of the two days is statistically significant;the comparison between the groups,the self-made prescription ointment group and the Erhuang Xinshang Zhitong ointment group are not significantly different from each other.Compared with the model group,the i NOS in the tissues of the two groups are not statistically significant.The expression of is significantly decreased,which is statistically significant(P<0.05).(6)The activity of c NOS in injured tissues: There was no statistical significance in the comparison of rats in each group,whether between groups or within groups.Conclusion: The self-made prescription ointment can significantly improve the symptoms and signs of acute soft tissue injury in rats,and promote the recovery of injured limbs;the self-made prescription ointment has the same effect as Erhuang Xinshang Zhitong ointment in the treatment of acute soft tissue injury in rats;self-made prescription ointment The mechanism of treatment of acute soft tissue injury may be related to inhibiting the expression and activity of i NOS in the injured tissue and reducing the overproduction of NO in the injured tissue.
