Effects Of Chubi Jianxie Decoction On Serum IL-1? And TNF-? Levels In Rats With Knee Osteoarthritis

Objective: Chu bi Jian xi decoction is an empirical prescription based on long-term clinical experience and clinical efficacy of Professor Wang Yong,a famous traditional Chinese medicine in Hunan Province.In order to further study the effect of Chu bi Jian xi Decoction on serum IL-1?and TNF-? levels in rats with drug-induced knee osteoarthritis,and further clarify the partial mechanism of Chubi Jianxi Decoction in the treatment of knee osteoarthritis.Methods: 70 SPF male rats aged 7 weeks were randomly divided into 7groups: A(Tenghuangjiangu group),B(high concentration Chu bi Jian xi Decoction group),C(medium concentration Chu bi Jian xi Decoction group),D(low concentration Chu bi Jian xi Decoction group),E(model group),F(sham operation group),G(blank group),with 10 rats in each group.Among them,A,B,C,D,E five groups used joint cavity injection of Sodium iodoacetate to induce drug induced knee osteoarthritis(KOA)rat model,F group used the same way injecting normal saline into the joint cavity,G group did not deal with it.The drug was given 14 days after modeling,Group A,B,C and D were given Tenghuangjiangu and Chu bi Jian xi Decoction at high,medium and low doses respectively,E.Group F was given the same volume of normal saline,twice a day for 14 days.The blank group was fed normally without any treatment.After 14 days of administration,the rats were sacrificed to prepare the knee joint specimens,the morphological and histological changes of knee cartilage were observed,and the effects of different drugs on the contents of IL-1 and TNF-? in serum were observed.Results : 1.Chu bi Jian xi decoction can effectively reduce the local swelling of knee joint in koa rats,and there is no significant difference between different concentration groups.2.Chubi Jianxi decoction can effectively improve the pathological damage of cartilage in koa rats,and its curative effect may increase with the increase of dosage in a certain range.3.Chubi Jianxi decoction can effectively reduce the contents of IL-1?and TNF-? in serum of KOA rat and its curative effect may increase with the increase of dosage in a certain range.4.The effect of Chu bi Jian xi Decoction on repaircartilage,reducing IL-1 and TNF-? in serum of KOA rats was similar to that of Tenghuangjiangu.Conclusion: Chu bi Jian xi decoction can effectively reduce the contents of IL-1? and TNF-? in the serum of model rats with KOA,which may be part of the mechanism of Chubi Jianxi Decoction in the treatment of knee osteoarthritis.
