Effect Of Tongluozhibi Decoction On The Content Of IL-1? And TNF-? In Articular Cartilage And Serum In Rabbit Knee Osteoarthritis Model

Purpose:Through the animal experiment of rabbit knee osteoarthritis model,it was confirmed that the treatment effect of "Tongluo Zhibi Decoction" on KOA and its mechanism for the effect of KOA was the clinical application of Tongluo Zhibi decoction in the treatment of knee osteoarthritis data support.Medthod:Forty healthy New Zealand rabbits were randomly divided into Tongluo Zhibitong group,rattan huangjian bone pellet group,model group and blank group.In addition to the blank group,the other groups were treated with modified Hulth method(Xiong Yuan,rabbit knee osteoarthritis model preparation and identification,experimental animal science,2013,30(3),31?32.)Preparation of rabbit knee osteoarthritis model After treatment,Tongluo Zhibi Decoction group was treated with Tongluo Zhibi Tang Granule granules,and the rats of Guoguangjuebao group were treated with the solution of Gushanjianbiao pill,the blank group,the model group were treated with normal saline,To determine the dose according to Chen Qi"Chinese medicine pharmacological research method(second edition)",the drug component dose by adult clinical daily dose of 5 times to adult weight to 60 kg as the standard,the daily dose per rabbit body weight For:(120ml/60kg)X rabbit body weight(kg)calculated.Once a day,continuous gavage 8 weeks.After 8 weeks of radiotherapy,the structure and morphology of chondrocytes were observed by HE staining,and the cartilage Mankin's score was performed.The levels of IL-1? and TNF-? in serum were measured by enzyme-linked immunosorbent assay(ELISA).Rsults:a.The articular cartilage surface and the edge of the joint were light,the color was fresh,no osteoporosis and cartilage damage.The chondrocytes were arranged neatly under the light microscope.In the model group,the articular cartilage was obviously damaged,the cartilage was white,and the cartilage was localized.The cartilage was thickened at the edge of the joint.The chondrocytes were reduced by light microscopy,and the cell cluster was lost.There was no significant difference between Tongluo Zhibi Decoction group and Tenghuang group.The degree of cartilage destruction and the level structure under light microscope were better than those in the blank group.b.Cartilage Mankin's score:Tongluo Zhibi decoction group and rattan huangjian bone group score score was significantly lower than the model group(P<0.05);Tongluo Zhibi decoction group,rattan huangjian bone group and model group and blank Group,the first three scores were high(P<0.05).c.ELISA test:the content of IL-1? in the serum and articular fluid of collateral-dredging Arthralgia-treating decoction group were markedly lower than that of model group,although the difference between the positive control group and it was not significant,the latter was lower than the former in terms of content of IL-1?.c.ELISA test:?IL-1?content:The levels of IL-1? in serum of Tongluo Zhibi decoction group and rattan huangjian bite group were significantly lower than those in model group(*P<0.05).?TNF-a content:The serum TNF-a level in the model group was significantly higher than that in Tongluo Zhibi Decoction group,rattan huangjian bone pellet group and blank control group(*P<0.05),the difference was statistically significant,the levels of serum TNF-a in Tongluo Zhibi decoction group and rattan huangjian pill group were higher than those in blank control group.Conclusiona.Modified Hulth method can successfully simulate knee osteoarthritis model.b.Inflammatory factors IL-1? and TNF-? cause knee osteoarthritis by inducing damage to articular chondrocytes.c.Tongluo Zhibi Decoction can significantly reduce serum IL-1? and TNF-? in rabbit knee osteoarthritis,by inhibiting IL-1? and TNF-? levels,delay the progress of KOA disease,play its role in the treatment of KOA.
