Objective:To observing the effect of guan j i e ruanjian zhitong tablet on IL-1 and TNF-? in serum and articular fluid of rabbit model of knee osteoarthritis,explore the mechanism of the treatment of knee osteoarthritis by guanjie ruanjian zhitong tab let.Methods:40 ordinary New Zealand rabbits were selected.Using random number table method,they were divided into blank group,modelgroup,sodium hyaluronate group,guanjie ruanjian zhitong tablet low-dose group(low-dose group)and guanjie ruanjian zhitong tab let high-dose group(high-dose group),with 8 rats in each group.Except blank group,the knee osteoarthritis model of rabbits in the other 4 groups was established by Plaster and wire fixation in flexion position.Medical intervention was given immediately after the completion of the modeling.One week after gavage,serum from abdominal aorta and knee joint fluid were collected,and the contents of IL-1 and TNF-? in serum and joint fluid were detected by ELISA.The internal femoral condyle cartilage of the left posterior knee of the rabbit was completely removed surgically,and the internal and external femoral condyle of the rabbit was observed by the naked eye.The histological morphology of the articular cartilage of the internal femoral condyle of each group after HE staining was observed under light microscope,and Mankin's score was performed.Results:1.The gross observation of the cartilaginous cartilage of the internal and external femoral condyle:the blank group had bright red color,smooth surface and smooth articular surface;In model group,cartilage lost luster,synovium thickened,articular surface was not smooth,joint deformation and local uneven,cartilage defects,partial subchondral bone and osteophyte format ion were visible.In the sodium hyaluronate group,the cartilage was less lustrous,the articular surface was slightly uneven,the synovial membrane was thickened,the cartilage defect and osteophyte formed were better than those in the model group.The situation of the low-dose group and the high-dose group was roughly the same,and the overall situation was between the blank group and the sodium hyaluronate group.2.The contents of IL-1 and TNF-? in serum and articular fluid of model group,low-dose group,high-dose group and sodium hyaluronate group were higher than those of blank group(P<0.05).The contents of IL-1 and TNF-? in serum and articular fluid of model group were consistent with the pathological signs of knee osteoarthritis,and were higher than those of low-dose group,high-dose group and sodium hyaluronate group(P<0.05).The content s of IL ? 1 and TNF-? in serum and articular fluid of sodium hyaluronate group were higher than those of high and low dose groups(P<0.05).There was no significant difference in the contents of IL-1 and TNF-? in serum and articular flu id between high-dose group and low-dose group(P>0.05).3.Mankin' s scores of model group,low-dose group,high-dose group and sodium hyaluronate group were higher than those of blank group(P<0.05).Mankin's scores of sodium hyaluronate group,high-dose group and low-dose group were lower than those of model group(P<0.05).Mankin's scores in the high-dose and low-dose groups were lower than those in the sodium hyaluronate group(P<0.05),and there was no statistical significance in the Mankin' s scores between the high-dose and low-dose groups(P>0.05).Conclusion:1.Guanj ie ruanjian zhitong tablet can reduce the content of IL-1 TNF-? in serum and articular fluid of rabbit knee osteoarthritis model,protect the cartilage,and has the effect of treating rabbit knee osteoarthritis.Equivalent dose,no need for high dose;The mechanism may be related to inhibiting the expression of IL-1,TNF-? and other inflammatory factors.2.The knee osteoarthritis model of rabbits can be established by Plaster and wire fixation in flex ion position.3.The effect of joint soft and strong pain killer tab let on rabbit knee osteoarthritis is better than that of sodium hyaluronate injection alone.
