Objective:To determine the status of HPV infection and HPV genotypes in Tibetan women in Diqing Prefecture,Yunnan Province;to understand the structure and composition of microorganisms in the reproductive tract of Tibetan women in Diqing Prefecture by analyzing the results of cervical and vaginal microbial sequencing;and to explore the correlation between microorganisms in reproductive tract and HPV infection.Objects and methods:Collect the women in Diqing Prefecture of Yunnan Province who participated in the study voluntarily by the people’s Hospital of Diqing Autonomous Prefecture in September 2020,collect basic information(including height,weight,age,etc.);and take three cervical swabs,and vaginal swabs,select Tibetan cases,and select appropriate cases according to the inclusion and exclusion criteria.The HPV genotypes of the samples collected from selected cases were detected,and the genomic DNA of the samples was extracted.The extracted DNA was amplified by universal primers of bacterial 16srRNA gene,and the bacterial community structure was analyzed by Illumina Miseq high-throughput sequencing.Analysis method:(1)SPSS26.0 software was used for data analysis.The measurement data were expressed by(±s),the independent sample t-test was used for the comparison between groups,the counting data was expressed as n(%),and the chi-square test or Fisher exact test was used for the comparison between groups.(2)The data returned by sequencing are analyzed and processed according to the software package of MiSeq SOP,namely Mothurv1.42.0[1].The source of database sequence and classification information is SILVASSURefdatabasev132[2].Compare the sequence similarity,select the similarity greater than 97%to divide the operable taxon OTU,and calculate the Alpha diversity index of each sample in turn.The OTU with high abundance and representative was selected for analysis,and the distance matrix and clustering of all sample sequences were calculated.Based on the calculated results,principal coordinate analysis PCoA was carried out,and the OUT with higher abundance and more representative was selected for comparative analysis.Result:Finally,there were 39 cases of Tibetan women,of which 16 cases of HPV+(positive for cervical HPV infection)accounted for 41.03%of the total,23 cases accounted for58.97%of the total number of people.According to the cervical HPV infection,the patients were divided into HPV+and HPV-groups,and the general data of the population were analyzed.It was found that there were significant differences in constipation,body weight and body mass index(Bodymassindex,BMI)between HPV+and HPV-groups,that is,the incidence of constipation in HPV+and HPV-positive group was significantly lower than that in HPV-negative group,and the body weight and BMI of HPV+were significantly lower than those of HPV-.The genotypes of HPV infection in Tibetan women included 16 genotypes,including 1 case with multiple genotypes,genotypes 16,18,51 and 67,and the others were single genotypes with 16genotypes.At the same time,the situation and genotype of vaginal HPV infection were also detected.There were 9 cases of vaginal HPV+and 30 cases of vaginal HPV-.The genotypes of HPV in cervix and vagina infection did not completely overlap with the cases of vaginal infection.The analysis of reproductive tract microbial diversity index showed that there were significant differences between cervical microorganisms in HPV+cases and vaginal microorganisms in HPV+and HPV-cases.Based on the diversity analysis of the microbial community in the reproductive tract,it was found that the cervical microbial community abundance of HPV+cases was significantly different from that of vaginal microflora and cervical microbial community of HPV-cases.There was no significant difference in the composition of microorganisms in vaginal and cervical samples of HPV+and HPV-cases by PCoA analysis.LEf Se analysis showed that there were differences in microbial community abundance in different parts of reproductive tract under different conditions of HPV infection.
