| Charpterl Prevalence Characteristics of Cenical Human Papillomavirus(HPV)Genotypes in Guangxi;data from One Medical CenterObjectives The prevalence of HPV infection and HPV genotypes varies in different regions.However,epidemiological characteristics are the basis of prevention and treatment.This study conducted a ten-year survey in a health center,to estimate the prevalence characteristics of HPV genotypes.Methods By using polymerase chain reaction(PCR)amplification and nucleic acid molecular hybridization,the HPV genot>pes were detected from 77,756 females who were patients of the Department of Obstetrics and Gynecology and those who visited the Health Management Center for a physical examination between August 2011 and November 2020.The prevalence,genotypes,age-related HPV infections,as well as chronological change of HPV prevalence and the HPV genotype distribution were analyzed.Results The overall prevalence of HPV infection was 21.14%(16,439/77,756).The HPV infection rate differed significantly between the patients of the Department of Obstetrics and Gynecology and the women who underwent a physical examination(22.98% versus9.88%,< 0.05).The prevalence rates of high-risk HPV,low-risk HPV,mixed HPV(mixed high-risk,and low-risk HPV infection),and mult^le HPV infections were18.96%(14,739/77,756),4.09%(3,178/77,756),1.90%(1,478/77,756),and 4.94%(3,838/77.756),respectively.The most prevalent genotypes were HPV 52,16.and 58.The age-associated HPV prevalence showed biniodal curves,with the first peak at<25 years and the second peak at >56 years.Conclusions This study provides baseline data on the HPV prevalence in the general female population of Nanning,Guangxi Province.Women <25 and >56 years old feced the greatest threat of HPV infection,and HPV 52,16,and 58 were the most common genotypes.Charpter2 Epidemiological characteristics of HPV infection in reproductive tract of middle-aged and elderly women in one medical center of Guangxi,ChinaObjectives To analyze the type and distribution characteristics of human papillomavirus(HPV)infection along with cervical cytology in middle-aged and elderly women in Guangxi,to provide a basis for the prevention and treatment of cervical cancer in elderly women.Methods 21 subtypes of HPV and cervical cytology of women over 45-year-old visit the First Affiliated Hospital of Guangxi Medical University from January 2019 to December 2020 were collected.They were divided into two groups by age.45-64 years old group and over 65 years old groiq).The HPV.HR-HPV.and multiple HPV infection prevalence were analyzed,as well as HPV genotypes,the age distribution of HPV infection rate,and cervical cytology.Results 6 657 eligible women were included,6 238 women were in the 45-64 years groiq),their HPV prevalence was 20.86%,while 419 women were in the over 65 years group,the HPV prevalence was 32.94%.The age-associated HPV prevalence showed rising curves before 70-year-old,peaked at the age groip of 70-74 years old groiQ).The most prevalent genotype was HPV52,the infection rate was 5.3%.followed by HPV16 and HPV 58.The majority cytology of HPV-positive elderly women was normal 8.7% of them were ASC-US,6.52% were HSIL,4.55% were LSIL,and 2.96% were ASC-H,1 of them was SCC,conpared to middle-aged women,abnormal cytology rates were higher,P<0.05.Conclusions HPV and HR-HPV prevalence of elderly women in a medical center of Guangxi were higher than middle-aged women The most prevalent genotype was HPV16 for elderly people,followed by HPV52 and HPV 58.Charpter3 Constituent ratio of human papillomavirus infection and the relation to aging in women from different ethnic groups in Guangxi areaObjectives To study the constituent ratio of human papillomavirus among women from different ethnic groins in the Guangxi area,and provide epidemiological data for the prevention and treatment of HPV infection.Methods Analyzed retrospectively HPV genotyping results of women from different ethnics groups,who visited the Department of Gynecology and Obstetrics of the First Affiliated Hospital of Guangxi Medical University from August 2012 to October 2020.They were divided into Han group,Zhuang group.PCR amplification was used to detect 21 HPV genotypes.Results 21304 HPV genotjping results were collected,in which 16390 were in the Han group,4346 were in the Zhuang groiq^.Overall,5591(26.96%)women were HPV positive.In the Han groip,4536(27.68%)women were positive,while in the Zhuang groi^).1055 women(24.28%)were positive,there was a statistical difference between them.The most common genotypes in both ethnics groups were HPV 16,52,58,18,and 53.HPV positive constiti^nt ratio in Zhuang groiqp rose with age,and peaked in the age group of over 65 years,while in Han women it presented a binx>dal curve,in which the first peak was in the age groip of younger than 20 years,and the second and highest HPV infection rate in the age group of over 65 years.In women with HPV positive,the proportion HPV16 and HPV 52 rose with age,while that of HPV 6 and 11 presented the opposite trend.Conclusion The HPV constituent ratio of Zhuang women was bwer than Han womea Zhuang women got the highest infection rate in the age groups of over 65 years.The five most common HPV genotypes were HPV 16,52,58,18,and 53.In women with HPV positive,the proportion HPV16 and HPV 52 rose with age,while that of HPV 6 and 11 presented the opposite trend.Purpose: This study investigated the role of Human umbilical cord-derived mesenchymal stem cell conditional medium(hUCMSC-CM)in relieving IUA,finding out whether Wnt/p-catenin signaling was involved,if so,e>q3lore the mechanism and determine the possible ligands,to provide new research targets for stem cell therapy,and the theoretical basis for the development of relevant targeted drugs.Methods: After being treated with hUCMSC-CM,the proliferation and migration of endometrial epithelial cells(EECs)were measured by CCK8 assay and Scratch assay.The activation of Wnt/p-catenin signaling was measured by Western blots,fluorescent staining,and TCF/LEF luciferase.Wnt inhibitor(XAV393)was used to inhibit the proliferation effect of UCMSC-CM in EECs.Wnt5 a expression in hUCMSCs was measured by Western blots,fluorescent staining,and Wnt5 a in hUCMSC-CM was detected by Elisa,to further clarify the mechanism.Results: hUCMSC-CM promoted proliferation and migration of endometrial epithelial cells(EECs),as shown by the CCK8 assay.The egression ofp-catenin,c-myc? and cyclin D1 increased in EECs after being treated with hUCMSC-CM.hUCMSC-CM was also found to promotedp-catenin delivery into nuclei by Western blot and fluorescent staining,meanwhile,the inhibitor(XAV393)can restrain this process and inhibit the proliferation effect of UCMSC-CM in EECs.Wnt5 a was detected in hUCMSCs and hUCMSC-CM,which might be a potential signal protein.Conclusion: This study demonstrated that hUCMSCs-derived conditioned medium promoted human endometrial cell proliferation through Wnt/p-catenin signaling,and Wnt5 a might be a potential activator.This would be one of the activating signal pathways in IUA’s MSCs-related treatment.Limitation:① It is preliminarily found that hUCMSC-CM can promote the proliferation and migration of human endometrial epithelial cells,but the analysis of endometrial cell cycle after hUCMSC-CM treatment is not conprehensive and lacks of strength?② Only one of the molecules involved in the catenin signaling pathway was detected,other proteins,Fzd receptor,LRP5 / 6,APC,CKla,and GSK-3P were not tested.(S)The relationshq)of Wnt5 a and Wnt signaling in endometrial epithelial cells has not been established.?
