Study On The Law Of Medication Use Of Professor Huang Guihua In The Treatment Of Peptic Ulcer

Objective: In order to inherit the academic thoughts and clinical experience of famous experts,Professor Huang Guihua’s prescriptions for peptic ulcer were excavated,and his clinical experience and academic thoughts were summarized.The core prescription drugs obtained by data mining were used to predict the relationship between active ingredients,disease targets and pathways,and explore the mechanism of action of core drugs on diseases.Methods:(1)According to the inclusion criteria and exclusion criteria,the medical records of peptic ulcer diagnosis and treatment in outpatient department of tutors were collected.After inputting the elements into excel,the frequency of drugs and the frequency of symptoms of patients were counted by traditional Chinese Medicine Inheritance calculation platform.The algorithm used association rules and K-means clustering.(2)After data mining,TCMSP,Gene Cards and OMIM were used to collect the active ingredients and potential targets of peptic ulcer.PPI network diagram was drawn.GO function and KEGG pathway enrichment were analyzed by Metascape.IGEMDOCK software was used for molecular docking.Results:1.Research results of Professor Huang Guihua’s medical records on peptic ulcerHerb-medicine information: a total of 88 herb-medicine,the frequency of1789,high-frequency herb-medicine are Atractylodes macrocephala,Amomum villosum,cardamom,fried licorice,etc.The most frequently using are tonifying deficiency medicine,dehumidification medicine and anti-eating medicine.The frequency of Qi tonifying drugs was the highest,followed by Yang tonifying drugs.Most of the drugs are sweet and warm.High frequency drug pairs included Sand kernel + nutmeg,sand kernel + atractylodes,atractylodes +nutmeg,atractylodes + licorice,sand kernel + licorice.Fennel→ Atractylodes,Atractylodis Rhizoma→ Pogostemon cablin,Psoralen→ Atractylodes,the confidence of three combinations was 1.Symptom tongue and pulse information: 117 clinical symptoms and tongue and pulse conditions were collected from the standardized medical records,with a total frequency of 1067 times.The high frequency symptom combinations were dry mouth + bitter mouth,acid regurgitation + belching,epigastric pain + pale tongue,pale tongue + white fur,etc.the 17 association rules,such as epigastric pain,bitter mouth,fecal rarity → dry mouth,pale tongue,belching,weak pulse → acid regurgitation,all had 1 confidence.In the drug cluster,Atractylodes macrocephala,cardamom,Rhizoma Pinelliae,Poria cocos,Amomum villosum and Codonopsis pilosula had the highest frequency of similar prescriptions.2.Network pharmacology prediction results of core prescriptionsThere are 64 potential intersection targets of drug active ingredients and diseases;Jun,AKT1,TNF in PPI network are highly connected targets;their biological processes include active oxygen metabolism,reaction to lipopolysaccharide,oxidative stress,etc.The enrichment of related molecular functions mainly includes cytokine receptor binding,heme binding,antioxidant activity,etc.MMP9 and quercetin are the best binding targets and compounds in molecular docking.Conclusions:1.Conclusion of Professor Huang Guihua’s research on medication regularity of peptic ulcerProfessor Huang Guihua diagnosed and treated peptic ulcer,carefully examined the cold and hot Yin and Yang,treated according to the syndrome,taking into account the regional hot and humid climate and modern environmental impact.The dampness pathogen was heavy,first removing dampness,then strengthening the spleen and protecting the kidney,strengthening the foundation and cultivating the yuan.In the treatment,he could predict the change of the disease,dispatch the prescription to attack and defend,and at the same time,he was good at the combination of drugs,so as to achieve synergy.2.Conclusion of network pharmacology analysis of core prescriptionsData mining medication rules to get Professor Huang Guihua’s core drug prescriptions(Atractylodes macrocephala,cardamom,ginger pinellia,Poria cocos,Amomum villosum,Codonopsis pilosula)for the treatment of peptic ulcer,using network pharmacology tools to predict the drug components that play an important role,visually showing the mechanism of the target on the pathway.Among the core prescriptions represented by Sijunzi Decoction,quercetin,luteolin,baicalein and other traditional Chinese medicine components can play the role of antioxidant,anti-inflammatory,repairing gastric mucosa and participating in immune regulation by affecting PPARG,MMP9,VEGF,PTGS2 and other key targets,combined with go function enrichment analysis and KEGG pathway enrichment analysis,These potential targets are closely related to the metabolic process of reactive oxygen species,oxidative stress response and response to toxic substances in biological function and signal pathway enrichment,so as to play a therapeutic effect.
