Exploring The Core Compatibility And Mechanism Of Treating Peptic Ulcer Based On Data Mining And Network Pharmacolog

Purpose:1.The first TCM prescriptions for the treatment of peptic ulcer patients in the Department of Spleen and Stomach,Affiliated Hospital of Liaoning University of Traditional Chinese Medicine from September 2019 to September 2021 were included and screened.2.To explore the clinical medication rules of TCM in the treatment of peptic ulcer through data mining,and explore the common compatibility of peptic ulcer treatment.3.Retrieve and predict the active ingredients,action targets and signal pathways of the compatibility of common Chinese medicines for the treatment of peptic ulcer through network pharmacology.Taking "drug-active component-target-signal pathway" as the main line,the mechanism of action of peptic ulcer treatment was explored from multiple perspectives and levels,providing reference for the development and research of new drugs.Materials and Methods:1.According to inclusion and exclusion criteria,TCM prescriptions for peptic ulcer treatment in department of Spleen and Stomach,Affiliated Hospital of Liaoning University of Traditional Chinese Medicine from September 2019 to September 2021 were recorded and a database was established.2.WPS software was used to analyze the frequency,category,property,flavor and meridian of drugs.Then SPSS Modeler18.0 software was used for correlation analysis.Cluster analysis was performed by SPSS Statistics 27.0 software to obtain the law and common compatibility of Chinese medicine for clinical treatment of peptic ulcer.3.Search the active ingredients of the above "common compatibility" in TCMSP database,predict the target information,and establish the database of active ingredients and action targets.Potential targets of peptic ulcer were searched in Gene Cards and Disgenet databases.Venny2.1 software was then used to make the intersection between the target of "tangerine bark-bupleurum" and the potential target of peptic ulcer to obtain the common target of both and draw the Wayne diagram.The common targets were imported into the STRING11.0database to construct the protein interaction(PPI)network.Cytoscape MCODE plug-in was used to construct molecular interaction modules in PPI network,and Cyto NAC plug-in was used to screen key targets.The common targets were introduced into Metascape database for GO functional annotation analysis and KEGG pathway enrichment analysis.Finally,the network diagram of "common compatibility-active component-action target-signal pathway" was constructed by Cytoscape3.7.2 software.Results:1.A total of 106 prescriptions for the first treatment of peptic ulcer patients were collected after screening by inclusion and exclusion criteria.2.The law of traditional Chinese medicine for peptic ulcer showed that the frequently-used traditional Chinese medicine included tangerine peel,Bupleurum,licorice,Atractylodes atractylodes,Poria cocos,etc.The most commonly used drugs were tonifying drugs,regulating qi drugs,clearing heat drugs and relieving surface drugs.The order of drug property from high to low was warm,cold,flat,cold and hot.The top five were sweet,bitter,spicy,sour and light,and the top five were spleen,lung,stomach,liver and heart meridian.The cluster analysis was divided into five groups: The first group was ligusticum chuanxiong,Tubularum turmeric,White cardamom,Atractylodes atractylodes,Poncirus aurantii,Magnoliae Magnoliae,Bupleurum bupleurum,Plates and roots.The second category: coix seed,Yam,malt,amomum,astragalus,cinnamon,dried ginger,cassia twig,pinellia ternata,Perilla leaf;The third category was dandelion,Cuttlebone,Notoginseng,paeonia lactiflora,Rhizoma alba and liquorice.The fourth category was Codonopsis codonopsis,Cocos,Coptis chinensis,Scutellaria baicalensis and Angelica sinensis.In the association rules,tangerine peel and Bupelum were the most supported,which were the core compatibility for clinical treatment of peptic ulcer,followed by tangerine peel and licorice.3."Tangerine-Bupleurum" contains 19 active ingredients,224 targets,and 1024 potential targets for peptic ulcer.There were 92 common targets of "Tangerine-Bupleurum" and peptic ulcer.The main targets are AKT1,IL6,IL1 B,CASP3,JUN,MAPK3,PTGS2,EGFR,CXCL8 and PPARG.GO functional annotations and KEGG pathway enrichment analysis,according to the results of "dried tangerine or orange peel-radix bupleuri" in the treatment of peptic ulcer and oxidative stress,chemical stress,active oxygen metabolism,such as closely related to the biological process,and serine endopeptidase activity and hydrolytic enzymes,serine peptidase activity such as molecular function,involving multiple signaling pathways,Such as tumor necrosis factor,IL-17,HIF-1,C-type lectin receptor signaling pathway,etc.Conclusion:1.The TCM treatment of peptic ulcer mostly involves the traditional Chinese medicines such as warming and tonifying deficiency,regulating liver and qi,clearing heat and detoxifying.The meridian of peptic ulcer is also related to the spleen,stomach and liver meridian,so the main pathogenesis may be the weakness of spleen and stomach,the accumulation of evil heat and the obstruction of qi.In the association analysis,the first drug compatibility was "Tangerine peel-Bupleurum",which confirmed that the use of qi regulating drugs is indispensable in the treatment of peptic ulcer in TRADITIONAL Chinese medicine.2.According to the analysis of the active components,common targets and signal pathways of "Tangerine-Bupleurum" in the treatment of peptic ulcer,it is suggested that "Tangerine-Bupleurum" may play a role in the treatment of peptic ulcer by anti-oxidative stress,inhibiting the release of inflammatory factors and inhibiting cell apoptosis.
