Research On The Rule Of Prescription Medication For Coronary Heart Disease Based On Data Mining

Research Objective1 From Traditional Chinese Medicines Inheritance Assist System(TCMISS)software platform,the prescriptions of Traditional Chinese Medicines(TCM)and Chinese Patent Drugs treatment of coronary heart disease were collected to build database.Mining research methods such as complex system entropy cluster were used to analyze the utilizing frequency of the prescriptions drugs from the database.The results from these analyses will help clinical reasonable using of medicine and the development of new medicines.2 Using the network pharmacology methods to analyze the mechanism of the disease with the drug combinations.Research MethodsBased on TCMISS(TCMISS V2.0.1),The prescriptions of the traditional Chinese medicines and Chinese Patent Drugs treatment of coronary heart disease were collected to build database.Data mining research methods such as Apriori algorithm,which were based on association rules and complex system entropy cluster were used in the research to analyze the medicines data.And then analyze the mechanism of the drug combinations treatment based on the Cytoscape 3.5.1 software.1 The prescriptions of the traditional chinese medicines for coronary heart disease were collected to build database.Several important books are the source of these prescription,including?Taditional Chinese Medicines-Good Recipe For Coronary Heart Disease?,?Taditional Chinese Medicines Book-The Treatment Of Coronary Heart Disease?and?Taditional Chinese Medicines Book-Effect Of Coronary Heart Disease?.Data mining research methods such as Apriori algorithm,based on association rules and complex system entropy cluster were used in the research to analyze the utilizing frequency of the prescriptions drugs and compare the association between different medicines.Drugs with high the frequency of using and core combination were mined for further research.Based on above these results,the top five highest frequency drugs,including Salviae Miltiorrhizae Radix et Rhizoma,Chuanxiong Rhizoma,Poria,Astragali Radix and Trichosanthis Fructus,were selected to dig out the rules of how these drugs are used.The composition and combination of these drugs are the special points to explain.2 Based on the book?Chinese Patent Drugs?,the prescriptions of Chinese Patent Drugs for coronary heart disease were collected to build database.Association rules and complex system entropy cluster were used to analyze the utilizing frequency of these prescriptions drugs and association rules between different medicines.Drugs with high frequency of using and core drug combination were mined for further research.Based on these analyses,the Salviae Miltiorrhizae Radix et Rhizoma,Ginseng Radix et Rhizoma,Notoginseng Radix et Rhizoma were selected for exploring what the composition were and how to dominate with other drugs.3 The network pharmacology analysisBased on association rules and complex system entropy cluster,The drug combinations were obtained.Used chinese medicines for coronary heart disease were collected from?Taditional Chinese Medicines-Good Recipe For Coronary Heart Disease?,?Taditional Chinese Medicines Book-The Treatment Of Coronary Heart Disease?and?Taditional Chinese Medicines Book-Effect Of Coronary Heart Disease?.Based on the computed results from those 985 screened prescriptions,the five top frequently adopted drugs were Salviae Miltiorrhizae Radix et Rhizoma,Chuanxiong Rhizoma,Poria,Astragali Radix and Trichosanthis Fructus.The frequently adopted drug combinations were "Chuanxiong Rhizoma,Salviae Miltiorrhizae Radix et Rhizoma","Salviae Miltiorrhizae Radix et Rhizoma,Astragali Radix”,“Salviae Miltiorrhizae Radix et Rhizoma,Trichosanthis Fructus","Salviae Miltiorrhizae Radix et Rhizoma,Poria","Salviae Miltiorrhizae Radix et Rhizoma,Cinnamomi Ramulus".In the association rules with a confidence of over 0.7,the drug combinations included"Salviae Miltiorrhizae Radix et Rhizoma,Allii Macrostemonis Blbus->Trichosanthis Fructus","Allii Macrostemonis Blbus->Trichosanthis Fructus","Schisandrae Chinensis Fructus,Salviae Miltiorrhizae Radix et Rhizoma->Ophiopogonis Radix","Corydalis Rhizoma->Salviae Miltiorrhizae Radix et Rhizoma".36 core combinations and 18 new prescriptions were mined.(2)Study on the prescription rule of containing Salviae Miltiorrhizae Radix et Rhizoma:The five top frequently Miltiorrhizae Radix et Rhizoma?Chuanxiong Rhizoma and Carthami Flos were searched.Contains 136 compounds in Salviae Miltiorrhizae Radix et Rhizoma,189 compounds in Carthami Flos and 189 compounds in Chuanxiong Rhizoma.24 active molecules were selected in "Salviae Miltiorrhizae Radix et Rhizoma-Chuanxiong Rhizoma",The potential targets have 70.The five top of targets were NCOA1,F2,PTGS2,HSP90AB1,ADRB2.30 active molecules were selected in "Salviae Miltiorrhizae Radix et Rhizoma-Carthami Flos".The key targets were PTGS2 and ALOX5.Those active molecules may play roles through above targets.The related disease with those targets are cardiovascular disease,Cancer,Alzheimer disease,Asthma,Inflammation,Pain and so on.Research Conclusions1 The drugs of treating coronary heart disease mostly have the effects of activating circulation to remove blood stasis,tonifying qi and relaxing vein,reducing phlegm and resolving masses,which reflects a refined of the TCM principles to treat symptoms and root causes.This study has important directive for clinic,and TCM inheritance assist system software platform has valuable to mine the medication rules of TCM doctor.2 "Salviae Miltiorrhizae Radix et Rhizoma-Chuanxiong Rhizoma" and "Salviae Miltiorrhizae Radix et Rhizoma-Carthami Flos" have various chemical constituents,complex targets and involving broad disease.The active molecules may play roles through NCOA1,F2,PTGS2,HSP90AB1,ADRB2 and ALOX5.According to this study,Two-Drug Pairs can be used to do more research in cancer and inflammation disease.
