Study On The Medication Rules And Molecular Mechanism Of Modern Famous Traditional Chinese Medicine Practitioners In The Treatment Of Psoriasi

Objective:To explore the overall,southern and northern medication laws and core drug combination of modern famous and old traditional Chinese medicine doctors in the treatment of psoriasis,and analyze the molecular mechanism of the overall core drug combination of modern famous and old traditional Chinese medicine doctors in the treatment of psoriasis.Methods:(1)the data of 31 famous modern and old traditional Chinese medicine doctors prescriptions and medical records for the treatment of psoriasis recorded in ’Dang Dai Pi Fu Ke Zhong Yi Lin Chuang Jia Cong Shu’ were standardized,and the overall,southern and northern databases were established respectively;(2)The frequency statistics function of TCMISS V3.0 is used to make statistics on the use frequency of drugs in the prescription and the occurrence frequency of efficacy,four properties and five flavors and meridian tropism in drugs.Using the drug association rule function of TCMISS V3.0,the drug association degree and common drug combination are obtained by setting a reasonable number of support and confidence,and the overall,southern and northern core drug combinations are obtained through comprehensive analysis;(3)Using the cluster analysis function of TCMISS V3.0,the commonly used prescriptions of modern famous and old traditional Chinese medicine doctors in the treatment of psoriasis are classified by K-means algorithm,and various core drug combinations are obtained;(4)The TCMSP and TCMID databases are used to collect the compound components and action targets of the overall core drug combination of modern famous and old traditional Chinese medicine doctors.The Drugbank,Disgenet and Genecards databases are used to obtain the psoriasis targets.The intersection targets of compounds and diseases are taken.The "core drug combination-traditional Chinese Medicine-compound-target-psoriasis" network and PPI protein interaction analysis are constructed by Cytoscape and String,Metascape database was used for Go function and KEGG pathway enrichment analysis,and autodocktools were used to dock important compounds with core targets.Results:(1)Through the analysis of 215 prescriptions commonly used by modern famous and old Chinese medicine doctors in the treatment of psoriasis,it is found that the top 10 commonly used traditional Chinese medicine are Dihuang,Danggui,Gancao,Mu dan pi,Chishao,Tufuling,Danshen,Huangqin,Fangfeng,Baishao.The top 3 efficacy of traditional Chinese medicine are clearing heat,tonifying deficiency,promoting blood circulation and removing blood stasis.The top 3 properties are cold,warm and mild,the top 3 flavors are bitter,sweet and pungent,and the top 3 meridian tropism are liver meridian,heart meridian and lung meridian.The common core drug combinations of modern famous and old traditional Chinese medicine doctors composed of Dihuang,Chishao,Mudanpi,Gan cao and Tufuling were obtained.The involved prescriptions were divided into three categories.The core drug combinations of each categories were combination 1:Taoren,Honghua,Danggui,Chuanxiong,and Danshen;Combination 2:Dihuang,Mudanpi,Chishao,Jinyinhua,and Lianqiao;Combination 3:Gancao,Dihuang,Danggui,Baizhu and Fuling.(2)This paper analyzes the medical record of modern famous and old traditional Chinese medicine doctors in the treatment of psoriasis,involving 133 prescriptions in the South and 100 prescriptions in the North.The top 10 commonly used traditional Chinese medicine are Dihuang,Gancao,Mudanpi,Chishao,Tufuling,Baihuasheshecao,Huangqin,Shuiniujiao,Mai dong and Zicao in the South,Dihuang,Chishao,Mudanpi,Tufuling,Gancao,Jinyinhua,Baimaogen,Lianqiao,Zicao,and Danggui in the North;The top 3 efficacy of traditional Chinese medicine are clearing heat,tonifying deficiency and promoting blood circulation and removing blood stasis in the South,clearing heat,tonifying deficiency and promoting water and dampness in the North;the properties from high to low,the South is cold,flat,warm,cool and hot,and the North is cold,warm,flat,cool and hot;The flavors from high to low in the South and North are bitter,sweet,pungent,salty and sour;The top 5 meridian tropism are liver meridian,lung meridian,heart meridian,stomach meridian and spleen meridian in the South,liver meridian,stomach meridian,lung meridian,heart meridian and spleen meridian in the North.The southern core drug combination composed of Dihuang,Gancao,Mudanpi,Chishao,Tufuling,Baihuasheshecao,Shuiniujiao,Zicao and Danggui,and the northern core drug combination composed of Dihuang,Chishao,Mudanpi,Tufuling,Gancao,Jinyinhua,Bai maogen,Lianqiao and Banlangen were obtained.(3)Through the analysis of the overall core drug combination of modern famous and old traditional Chinese medicine doctors in the treatment of psoriasis,184 active compounds were obtained,including quercetin,kaempferol,baicalein,etc.236 targets of the core drug combination,1011 targets of psoriasis and 100 intersection targets were obtained.The core targets were TNF,IL6,IL1B,etc.Go analysis results mainly include biological processes such as response to xenobiotic stimulus,response to hormone,response to inorganic substance,positive regulation of cell migration,positive regulation of cell motility;The results of KEGG analysis included such as pathways in cancer,lipid and atherosclerosis,AGE-RAGE signaling pathway in diabetic complications,fluid shear stress and atherosclerosis,chemical carcinogenesis-receptor activation.Molecular docking results showed that quercetin,kaempferol and baicalein had good binding activities with TNF,IL6 and ILIB.Conclusion:The treatment of psoriasis by modern famous and old traditional Chinese medicine doctors mostly starts with blood,which is treated by clearing heat and activating blood,nourishing yin and dehumidification.The overall idea of doctors in the South and north is similar,but there are differences in medication,the South has strengthened the role of clearing heat,cooling blood and activating blood,and the North has strengthened the role of clearing heat,detoxification and dehumidification.The overall core drug combination in this study contains active compounds such as quercetin,kaempferol and baicalein,which can play a therapeutic role by interfering with immune response and cell proliferation of psoriasis with targets such as TNF,IL6 and IL1B.
