| Objective: Through the preparation of turkish galls cream(TGC)with drug residue extract(DRE)as raw material,the composition of the cream was analyzed qualitatively and quantitatively,the quality standard of the cream was established,the antibacterial activity of the TGC was determined,so as to control the quality of the TGC,and the residual value of the drug residue was better developed and utilized.Methods: The best extraction technology of drug residue was determined by orthogonal design;The best prescription and molding technology of TGC was selected by looking up a lot of literature and designing orthogonal experiment;Total polyphenols in the DRE and TGC were determined by Ultraviolet and visible spectrophotometry(UV-vis);Qualitative and quantitative analysis and preliminary antioxidant activity of gallic acid(GA)and methyl gallate(MG)in DRE and TGC were carried out by High-performance thin-layer chromatography(HPTLC)scanning and Thin-layer chromatography(TLC)autography;Qualitative and quantitative analysis of GA,MG and ellagic acid(EA)were carried out by High performance liquid chromatography(HPLC);In accordance with the national standard of the people’s Republic of China GB-15979-2002,the inhibitory effect of TGC on Staphylococcus aureus(SA),Escherichia coli(EC)and Candida albicans(CA)was determined by the test method of bacteriostatic property of dissoluble antibacterial(bacteriostatic)products,and the inhibitory effect of DRE and TGC on SA and EC was determined by agar drilling method and wet paper method.At the same time,the minimum inhibitory concentration(MIC)and the minimum bactericidal concentration(MBC)of the residue extract to SA and EC were determined;At 0,3,6,9,12 months,the characters of TGC were observed and recorded,including color,odor,texture,stratification,etc.At the same time,the content changes of GA,MG and EA were monitored,so as to investigate the stability of TGC.Results: The best extraction process of the drug residue was a material-liquid ratio of 1:10,60% ethanol,reflux at 85℃ for 30 minutes,and reflux twice;The best prescription and best molding process of TGC were 12 g stearic acid,3.5 g glyceryl monostearate,0.1 g paraben ethyl ester,1 g petrolatum,6 g liquid paraffin,5 g glycerol,0.4 g triethanolamine,1 g of DRE,adding water to 100 g,emulsification temperature 80℃,stirring time 30 minutes,stirring speed 800 r/min;The total polyphenol content of the three batches of DRE and TGC were in the range of476.29±12.4-565.34±3.94 mg/g、7.14±0.06-7.98±0.08 mg/g,respectively;In HPTLC,the thin layer plate was a polyamide film,and the developing agent was glacial acetic acid:methanol: ethyl acetate: formic acid(10: 6: 2: 1,v/v).The scanning wavelength was 280 nm.The retardation facter(RF)of the GA was 0.61±0.05,the RF of the MG was0.77±0.05,and the contents of GC and MG in the DRE were 91.91±3.37-129.3±5.29 mg/g,34.66 ± 2.93-48.10±3.89 mg/g,respectively.The content of GC and MG in the TGC was1.05±0.03-1.28±0.08 mg/g and 0.30±0.05-0.61±0.07 mg/g,respectively.Both GC and MG had certain antioxidant activity,and the antioxidant activity of GC was greater than that of MG;The contents of GC,MG and EA in DRE were determined by HPLC at140.09±0.99-167.02±2.68 mg/g,9.13±0.08-10.6±0.05 mg/g,44.23 ±0.25-51.46 ± 0.04mg/g,respectively.The contents of GC,MG and EA in TGC was in the range of2.31±0.01-2.76±0.01 mg/g 、 0.56±0.01-0.64±0.01mg/g 、 0.13±0.00-0.18±0.00mg/g,respectively;The results of in vitro bacteriostatic experiments showed that DRE and TGC had certain inhibitory effects on SA,EC,and CA.The results of long-term stability experiments showed that TGC was stable within 12 months.Conclusion: In this study,TGC was prepared by using drug residue as a raw material,and a qualitative and quantitative analysis method for the ingredients in the cream was established,and the inhibitory effect of TGC on SA,EC,and CA was initially determined.This experiment provides new ideas for the secondary development and utilization of medicinal residues,which can promote the development of new products,create more benefits for enterprises,and also contribute to environmental protection. |
