Eucommia ulmoides(Eucommia ulmoides Oliv.)is a perennial deciduous tree of Eucommia ulmoides,which is used as a dried bark.It has a long history of use and is contained in the Shennong Bencao Classic.Sweet,warm,with liver and kidney,strong bones and muscles,the effect of fetus,medicinal value is very high.However,the quality control method of Eucommia ulmoides is not perfect at present,and only the content of rosinol diglucoside is used as the quality control method of Eucommia.As a commonly used traditional Chinese medicine and health care products,it has been reported in the literature that Eucommia ulmoides has potential genotoxicity,in order to improve the safety evaluation data of Eucommia.In this study,the chemical constituents of Eucommia ulmoides were qualitatively quantified,and the safety of Eucommia ulmoides was initially explored.The details are as follows:1.Qualitative and quantitative analysis of main chemical constituents of Eucommia ulmoides based on LC-MSA qualitative and quantitative analysis method for the main components of Eucommia ulmoides Oliv.was established by ultra performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS).UPLC-Q-Exactive orbitrap high-resolution mass spectrometry and negative ion detection mode were used to identify 98 compounds,including 38 lignins,13 iridoids,9 flavonoids,and phenylpropanoids.15 phenols and 7 phenols were used.The UPLC-MS/MS method and negative ion MRM detection mode were used to analyze rosinol diglucoside,rosinol monoglucoside,syringin diglucoside and peach in Eucommia ulmoides Oliv.The content of seven main components of saponin,genipin,geniposide,and stearyl.The linear relationship of the components measured in the concentration range examined was good(R~2>0.9915),the precision RSD<3.42%,the reproducible RSD<4.32%,and the recovery rate(n=9)were 91.04%?108.15%,and the RSD was smaller than 4.89%,the results show that the method has high sensitivity and good reproducibility,and the analysis is fast,accurate and reliable.It can be used to qualitatively and quantitatively detect the main chemical components in Eucommia ulmoides Oliv.,and provide a reference for comprehensive evaluation of Eucommia quality.2.Study on single dose toxicity and repeated drug toxicity of Eucommia ulmoides extractAccording to the technical guidelines of single dose toxicity study,the maximum tolerance of Wistar rats to Eucommia extract is greater than 168g/kgBW.According to the technical guidelines for repeated drug toxicity test,the rats were given 13 weeks of drug administration.Wistar rats showed renal pathological changes at 11.2 g of crude drug/kgBW(equivalent to 84 times the clinical dose of human body).After 5 weeks of recovery,renal pathological changes returned to normal..At the dose of 56.0 g of crude drug/kgBW(equivalent to 420 times of human clinical dose),the drug was recovered for 5 weeks,and the pathological changes of rat kidney were still present.3.Study on genetic toxicity of Eucommia ulmoides extractAccording to the technical guideline of drug genotoxicity research,the results of bacterial mutation test showed that there was no obvious mutagenic effect on Salmonella typhimurium due to histidine deficiency within the dose range of 0.0325?22mg added to Eucommia ulmoides per dish.The micronucleus test of bone marrow cells in mice showed that there was no genotoxicity in Kunming mice within the dose range of 5.5?22.0g/kg BW of Eucommia ulmoides.The chromosome aberration test of mammalian cells in vitro showed that there was no obvious genotoxicity in the dose range of 2?20mg/mL given to Eucommia ulmoides.Genetic toxicity of Eucommia ulmoides The display is safe.
