Effects Of Turkish Galls Extract On TGF-β1、IL-6 Levels In Experimental IgA Nephropathy Rats

Objective: To investigate the effect of Turkish galls extract(TGE) on the the expression of Transforming growth factor-β1(TGF-β1), Interleukin-6(IL-6),Immunoglobulin A(IgA) in serum, urine and renal tissue of Immunoglobulin A nephropathy(IgAN) rats, and to further explore the possible mechanism. Methods: IgAN model rats successfully established for 12 weeks, and then they were randomly divided into IgAN model group, TGE 750 mg/kg group, TGE 150 mg/kg group, TGE 300 mg/kg group, and administrated intragastric gavage once a day for 4 weeks, normal group was added as control group as well, 20 rats in each group. At the end of week-12 and week-16,urinary red blood cells were respectively detected by urinary sedimserumalent microscopy and 24 hour urine protein were measured by BCA protein. At the end of week-16, serumal biochemical indexes were respectively detected by automatic biochemical analyzer,serumal and urinary TGF-β1, IL-6 and IgA levels were measured by enzyme linked immunosorbent assay(ELISA). And the expression of TGF-β1, IL-6 in renal were detected by immunohistochemistry. The deposition of IgA immune complex in the renal were observed by immunofluorescence assay. The renal pathological changes were evaluated with Oxford classification scoring system after HE, PAS staining.Renal morphology and ultrastructure were examined by electron microscopy. Results:(1) At the end of week-12,compared with the normal control group, urinary red blood cells and 24 hour urine protein increased significantly in all IgAN rats(P<0.05), suggesting that the IgAN models were successful.(2) At the end of week-16, compared with TGE 75 mg/kg group, serumal IL-6decreased most significantly in TGE 150 mg/kg group, the content of IL-6 in urine and renal tissue decreased most significantly in TGE 300 mg/kg group(P<0.05).(3) At the end of week-16, compared with TGE 75 mg/kg group, the content of TGF-β1 in urine, IgA in urine and renal tissue decreased most significantly in TGE 300 mg/kg group and TGE 150mg/kg group, the content of TGF-β1 in serum and renal tissue, IgA in serumdecreased most significantly in TGE 300 mg/kg group(P<0.05). Conclusions:(1) TGE can reduce urinary red blood cells and 24 hour urine protein in IgAN rats, decrease the expression of TGF-β1, IL-6 and IgA in in serum, urine and renal tissue.(2) TGE can inhibiti the proliferation of mesangial cells and tubular atrophy / interstitial fibrosis in IgAN rats, and has a certain curative effect on IgAN rats.(3) The mechanism of TGE postponing renal lesion may be associated with the down-regulation of the expression of TGF-β1, IL-6 and the inhibition of the deposition of IgA in the renal tissue.
