| Turkish galls is a dry cecidium of young wattle on Quercus infectoria Oliv.lt is formed by the parasitoid of young Cynips gallae-tinctoriae Oliv.The chemical constituents of turkish galls include tannins(50%-70%), galli acid(2%-4%) and resin etc.We studied the chemical constituents of it,and isolated five compounds by Sephadex LH-20 and MCI-GEL CHP -20P column chromatography.The compounds were testified by chemical and spetrum means such as UV, IR, 1H-NMR, 13C-NMR, 2D-NMR, ect,they are gallic acid ( â… ); m-digallic acid (â…¡); methyl gallate (â…¢); 1,2,3,6-tetra-O-galloyl- β -D- glucose (â…£); 1,2,3,4,6-penta-O- galloyl - β -D-glucose (â…¤) 。The extractive technology of Kui jiean enema was screened by means of Optimal experiment design,basing on the content of tannins, 1,2,3,6 -tetra-O-galloyl— β -D-glucose and extractum. The results showed that the best technology is 8 times volume water, three times boiling, each lasting 0.5 hour,concentrating soup to filtrate by plate-and-frame type filter, and add water to 1000ml, subpackage.Establish the quality criteria of Kui jiean enema. HPLC methold was applied to determinated the content of gallic acid and 1,2,3,6-tetra-O-galloyl-β -D-glucose in enema.The methold including linearity with range, detection limit, quantitation limit, recovery rate, precision, stability and repeatability.The methods are simple,rapid and accurate.Used accelerated test six months and long-run test nine months,we studied the stability of Kui jiean enema, and observed the character,relative density, microbiological margin and content of preparation. The results showed that the quality of preparation was stable. |
