Association Between MMP-3 Gene Polymorphism And Osteoarthritis In Han And Uygur Populations In Xinjiang

Objective:To explore the association between MMP-3 rs3025058,rs679620 gene polymorphisms and knee osteoarthritis((primary knee osteoarthritis,PKOA)in Uygur and Han osteoarthritis patients in Kashi,Tumushuke and Shihezi areas of Xinjiang.Methods:A case-control study was conducted in 111 Han OA patients,217 Han NOA subjects,105 Uygur OA patients and 201 Uygur NOA subjects in some areas of Xinjiang.Restriction fragment length polymorphism polymerase chain reaction(PCR-RFLP)was used to detect MMP-3 rs3025058 and rs679620 gene polymorphisms and analyze the correlation to Compare the differences in osteoarthritis and non-osteoarthritis between Han nationality and Uygur nationality Results:The genotypic distribution and allele frequencies of rs3025058 and rs679620 in Han and Uygur nationalities are in accordance with Hardy-Weinberg genetic equilibrium(P>0.05).There was significant difference in the distribution of5A/6A genotype and allele frequency of rs3025058 gene between Han and Uygur OA population(X~2=11.206,8.526,P<0.01).There was significant difference in5A/6A allele frequency of rs3025058 gene between OA group and NOA group in Uygur population(X~2=10.648,P<0.01),and compared with 6A allele,5A allele can increase the risk of Uygur OA(OR=1.793,95%CI:1.270～2.532).There was no significant difference in genotype and allele frequency of rs679620 locus between OA group and NOA group in Han nationality.(X~2=1.701,1.987,p>0.01).There was no significant difference in genotype and allele frequency of rs679620 locus between OA group and NOA group in Uygur population.(X~2=0.888,0.339,p>0.01).There was no significant difference in gene frequency and genotype between Han and Uygur OA groups.(X~2=0.241,0.557,p>0.01)。Conclusion:(1)MMP-3 rs3025058 gene polymorphism is associated with OA in Uygur population,and 5A allele may increase the risk of osteoarthritis in Uygur population.(2)there was no correlation between MMP-3 rs679620 and OA in Uygur and Han populations in Xinjiang.
