Objective:Osteoarthritis (OA) was considered a multi-gene expression was regulated by genetic disease. It has been demonstrated between the gene encoding a cytokine associated with OA susceptibility, interleukin (IL)-6gene also been implicated in cartilage degradation process. We in this case-control study to assess whether the risk of osteoarthritis in Han population could be IL-6174polymorphic loci influence.Methods:To evaluate the distribution of IL-6genotypes in112osteoarthritis patients and132age-and sex-matched healthy with its human and case-control study. By enzyme-linked immunosorbent assay (ELISA) study measured levels of serum IL-6, and the polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP method for the IL-6gene174C/G (rs16944) single nucleotide polymorphism loci analyzed, verified by sequencing digestion results.Results:The serum osteoarthritis of IL-6levels were significantly higher than those in healthy control group (t=-8.10, P<0.001), IL-6genotype distribution (1) patients with osteoarthritis:12GG,60GC,40CC (2) in the control group:9GG,33GC,90CC.CC genotype frequencies in the control group was significantly higher (P<0.02). Logistic regression analysis showed that, CC genotype proportion with OA (0.1e0.9P<0.04odds ratio0.4[95%confidence interval) is inversely proportional to the risk.Conclusion:Primary knee O A and important genetic variability genes encoding inflammatory cytokines, such as IL-6, may play an important role in the many processes pathophysiology of OA.
