Background:The incidence of knee osteoarthritis(KOA)is increasing year by year,but the exact cause and mechanism of knee osteoarthritis is not yet clear.Therefore,the study of the etiology and pathogenesis of knee osteoarthritis and the exploration of clinical treatment methods are still major problems and research hotspots for researchers.In recent years,with the deepening of research,knee osteoarthritis may be related to genetic factors.Objective:To explore the association between KOA and KOA TCM syndromes and single nucleotide polymorphisms of GDF5 gene.Methods:1 288 subjects were randomly selected to meet the standards(188 in the KOA group and 100 in the control group).The general data of the patients were recorded and the TCM syndromes were determined according to the diagnostic criteria of the TCM syndromes.2 Growth and differentiation factor 5(GDF5)plays an important role in the metabolism of articular cartilage.Studies at home and abroad have shown that multiple SNP sites in the 5'non-coding region of GDF5 gene rs224333,rs143383,rs224332 and OA The incidence is directly related,but the results obtained in different countries are different.Therefore,it is significant to select the Han nationality population in China to study the association of GDF5 gene single nucleotide polymorphism expression with the KOA and KOA TCM integer types.3 Blood samples were collected for DNA extraction,followed by single-base extension techniques and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry/mass spectrometry techniques for typing detection.Aggregate data,statistical analysis.Results:1 KOA was associated with gender and weight categories(P<0.05),but not with age;women(?~2=38.273,P=0.000<0.05)and obesity(P=0.009<0.05)all increased the risk of KOA.2 Hardy-Weinberg balance test,the SNP loci(P>0.05)selected in this experimental group are consistent with Hardy-Weinberg equilibrium.3 There was a correlation between female and KOA TCM syndromes(all P<0.05),and there was a correlation between obesity and KOA cold and dampness stasis(P<0.05).4 Alleles and genotypes of rs224333,rs143383 and rs224332 of GDF5 gene were not related to KOA and KOA syndromes(P>0.05).Conclusion:1 Women and obesity can increase the risk of KOA.2 There is a correlation between female and KOA TCM syndrome types,and females have the highest risk of KOA qi stagnation and blood stasis compared to men.There is a correlation between obesity and KOA cold and dampness.3 The single nucleotide polymorphisms of rs224333,rs143383,rs224332 in GDF5 gene may not be associated with the KOA and KOA syndrome types.
