| Allium Victorialis(AV)is rich in polyphenols and flavonoids,which can be used as natural raw materials for exogenous antioxidants.However,current studies on Allium Victorialis are mainly focused on the cultivation of Allium,the simple processing of marinated sauces,anti-inflammatory and other functional studies,and few studies on the purification and antioxidant of Allium Victorialis polyphenols.Therefore,in this experiment,Allium Victorialis was used as the raw material to obtain the crude extract of polyphenols of Allium Victorialis by water extraction method,and then the polyphenols in the crude extract were purified by macroporous resin and the antioxidant properties of the crude extract were studied in vivo.1.Purification of polyphenols in the crude extract of Allium Victorialis:taking the static adsorption rate desorption rate and dynamic adsorption rate desorption rate as indicators,through the comparison of five kinds of macroporous resin,AB-8 was finally determined as the optimal macroporous resin.The single factor experiment was used to obtain the optimal purification conditions for polyphenols of Allium Victorialis:pH=5,crude extract of Allium Victorialis polyphenols with a concentration of 0.9 mg/mL as the sample solution,the flow rate of the upper column was controlled at 1.0 mL/min,the eluent was 80%ethanol,and the eluent flow rate was 1.0 mL/min,the volume of sample loading was 90 mL,and the volume of eluent was 200 mL as the eluent condition.After purification,the purity of polyphenols rose from 2.74%to 36.09%,which increased by nearly 13 times.2.In Allium Victorialis antioxidant activity of polyphenols:the purified and crude extracts of polyphenols have higher reducing power to ferric ions,and strong scavenging ability to DPPH·free radicals and ABTS+ free radicals,and both of them are dose-dependent.The IC50 Allium Victorialis of DPPH·free radical scavenging rate of the purified polyphenol was 7.91 ug/mL,which was 0.56 times and 0.02 times of the IC50 Allium Victorialis of Vc and crude extract,respectively.The IC50 Allium Victorialis of ABTS + removal rate of the purified polyphenol was 72.87 ug/mL,which was about 2.07 times and 0.51 times higher than the IC50 Allium Victorialis of Vc and crude extract.The reduction rate of IC50 of the purified polyphenol was 171.59 ug/mL,which was about 2.18 times and 0.34 times higher than that of Vc and Allium Victorialis extract.3.Antioxidant activity in Allium Victorialis:mice were intraperitoneally injected with d-gal 35d to establish an oxidative damage model,followed by gastric intragastric test.Serum,liver and brain tissue were collected and stored in the refrigerator at-80℃ for 30 consecutive days.Then measure the relevant indicators.The results showed that compared with the D-g model group,the low-dose,medium-dose and high-dose groups showed that the organ coefficients of the mice in the low-dose,medium-dose and high-dose groups were significantly increased,and the liver and heart indexes in the high-dose group reached 46.08 and 4.73,respectively,which were the same as the Vc in the control group.The activity of Superoxide dismutase(SOD),Glutathione peroxidase(GSH-Px)and Catalase(CAT)in serum,liver and brain tissue of mice was significantly increased,and the content of malondiacede.mda was significantly reduced.The contents of 8-hydroxy-2’-desoxyguanosine(8-oh-d G)and 5-hydroxy-2’-deoxycotosine(5-oh-d C)in the serum and brain tissues of mice were significantly reduced.The content of 8-oh-d G in the high-dose group was 28.11 ng/L,which had recovered to the level of the normal group 25.84 ng/L.It significantly reduced the contents of tumor necrosis factor-beam(TNF-α)and interleukin-IL-6 in serum and liver tissues of mice(P<0.05).The liver tissue was also repaired to different degrees,and the condition of the high-dose group was basically restored to the normal group. |
