| Allium victorialis,is a perennial herbaceous plant belonging to the genus Allium in the Liliaceae family.It had dual edible and medicinal values.To provided an experimental basis for the further separation and the development of related products of Allium victorialis in the future.Therefore,in this experiment,Allium victorialis was used as the raw material,and the crude extract of the Allium victorialis was obtained by water extraction.The gradients of n-hexane(Hex),dichloromethane(DCM,ethyl acetate(EA)and n-butanol(BuOH)were used in sequence.After extraction,the contents of total phenols,total flavonoids,total saponins and total sugars of different polar extracts of Allium victorialis were determined,and the flavonoids of different polar extracts of Allium victorialis were analyzed by HPLC.The antioxidant properties of different polar extracts were investigated,and lipopolysaccharide(LPS)was used to induce RAW264.7 macrophages to establish an inflammation model.MTT method was used to detect cell viability,Griess method was used to explore the effect of different polar extracts of Allium victorialis on the release of NO from RAW264.7 cells induced by LPS,and ELISA assay was used to investigate the TNF-α effect of EA extract on RAW264.7 cells,IL-6 and IL-10 release.The research results were as follows:1.The yield of aqueous phase extract of gradient extraction was the highest,which is 77.75%,and the yield of DCM extract was the lowest,which is 1.35%.The total phenol and total flavonoid content of EA extract was the highest,the total phenol content was 334.20 mg/g,and the total flavonoid content was 127.27 mg/g.The total saponins content of the DCM extract was the highest,139.04 mg/g,and the total sugar content of the aqueous extract was the highest,259.30 mg/g.The different polar extracts of Allium victorialis all contained flavonoids.Astilbin,rutin and quercetin were the flavonoids with higher content in EA extracts,the contents were 8388.5μg/g,2245.82 μg/g and 1197.21 μg/g.Genistein,Yanagawa fish,quercetin and geraniol were the higher content of flavonoids in the DCM extract,the contents were 1172.65μg/g,818.64 μg/g,769.09 μg/g and 737.15 μg/g,respectively.2.Different polar extracts of Allium victorialis all showed different degrees of antioxidant properties,and present a dose-dependent.The IC50 values of the EA extract for scavenging DPPH free radicals and hydroxyl free radicals were the smallest,which were 0.153 mg/mL and 0.387 mg/mL,respectively,with the strongest reducing power,that was,the EA extract had the most significant antioxidant effect.Total phenols and total flavonoids were the material basis for the antioxidant activity of Allium victorialis.3.The different polar extracts of Allium victorialis could significantly inhibit the inflammatory response of RAW264.7 macrophages induced by LPS.The different polar extracts of Allium victorialis(100,300,500 μg/mL)could effectively inhibit the release of NO from macrophages,and there was a dose-dependent.The EA extract could inhibit the release of TNF-α and IL-6 from RAW264.7 macrophages.At the same time,it could effectively treat inflammation by promoting the secretion of IL-10. |
