Objective: To investigate the effect of edaravone combined with Danhong injection on the expression of mitochondrial autophagy protein SIRT5 in rats with acute cerebral infarction.Methods:The rats were divided into the following groups according to the random number table: sham operation group,model group,edaravone group,Danhong group,edaravone combined with Danhong group(combined administration group),Each groupwas divided into 4 subgroups at different time points(1d,3d,5d,7d),and each group received intraperitoneal injection of corresponding drug / d for 1 d,3 d,5 d and7 d respectively.The rats in the model group and the sham operation group were given equal volume of normal saline respectively.Then the brain tissues were decapitated at the corresponding time point.The neurological deficits The cerebral infarction area,brain water content and transmission electron microscope were used to observe the autophagy in rats.Immunohistochemistry was used to observe the morphological changes of hippocampus and the expression intensity of SIRT5 in rat hippocampus.The expression of SIRT5 was detected by q PCR.Results:The neurological deficit scores,brain water content and infarct volume in model group were higher than those in sham operation group on each day(P <0.05).Compared with model group,edaravone and Dan Compared with the model group,the scores of neurological deficit,brain water content and cerebral infarction in the red group and the combination group were significantly lower than those in the model group(P <0.05).Compared with the combination group,edaravone and Danhong group Neurological deficit score,brain water content,cerebral infarction volume difference is large,there was a significant difference(P <0.05).The mitochondrial auto Phagosomes were observed in the neurons of the sham operation group.The ex Pression and content of SIRT5 in the sham operation group were not significantly different(P> 0.05).The model The expression and content of SIRT5 in each group at each time point after cerebral infarction were higher than those in the control group,especially at 7th day(P <0.01).The combination group and Edaravone,Danhong Compared with each time Point,the ex Pression intensity and content of SIRT5 had statistical significance(P <0.05).Conclusion:Edaravone combined with Danhong injection can improve the neurological deficit in rats with acute cerebral infarction,reduce cerebral infarct volume and cerebral edema,and reduce nerve cell damage.The mechanism may be related to the regulation of SIRT5 protein expression and downregulation of mitochondrial autoantibodies.
