Objective:To observe the effects of Danhong Injection on acute cerebral infarction in rats,with the Neurological Severity Scores,expression of VEGF,MVD and changes in the volume of cerebral infarction,and to explore the mechanism of Danhong Injection on angiogenesis after cerebral infarction.Methods:100 healthy male SD rats of SPF grade,we were randomly selected20 rats as blank group,20 rats as sham operation group,the remaining rats were modeled by using the suture method,after the successful modeling,they were randomly divided into three groups:model group,Danhong treatment group,Butylphthalide treatment group,20 rats each,each group has two time points:7d and 14 d.Danhong treatment group received Danhong injection 4.2ml/kg intraperitoneal injection,and Butylphthalide treatment group was given butylphthalide injection 10ml/kg intraperitoneal injection,others were given saline.the rats were sacrificed at 7d and 14 d after the NSS was evaluated,and then,VEGF and MVD were measured by immunohistochemical,and the volume of cerebral infarction was measured by TTC staining.SPSS 22.0 software was used for statistical processing.Results:1.There were no neurological deficit in blank group and sham operation group rats,there were difference in NSS among model group,Danhong treatment group and Butylphthalide treatment group(P > 0.05);no neurological deficit was improved in 14 d than 7d of the three groups has statistically significant difference(P< 0.05).2.The expression of VEGF at each time point of blank group and sham operation group is rare,and had no significant difference between the twogroups(P > 0.05);model group,Danhong treatment group and Butylphthalide treatment group at each time point,the number of VEGF positive cells was significantly higher than the blank group and sham operation group(P < 0.05);the expression of VEGF on model group was increased after 7d,and then it tended to be stable;compared with the model group,Danhong treatment group and Butylphthalide treatment group at each time point,the expression of VEGF was significantly increased(P < 0.05),while the expression of VEGF has no difference between the two groups(P> 0.05).3.There were a small amount of blood vessels in blank group and sham operation group,there had no significant difference between two groups(P > 0.05);model group,Danhong treatment group and Butylphthalide treatment group at each time point,the MVD were higher than the blank group and sham operation group(P < 0.05),the MVD of 14 d was significantly less than the 7d among three groups(P< 0.05),compared with the model group,the MVD increased significantly in Danhong treatment group and Butylphthalide treatment group(P < 0.05),but there was no significant difference between Danhong treatment group and Butylphthalide treatment group(P> 0.05).4.There was no infarct in blank group and sham operation group,white infarcts were found in the right cerebral hemisphere of the rats in model group,Danhong treatment group and Butylphthalide treatment group.Compared with model group,the volume of cerebral infarction in the group of Danhong treatment group and Butylphthalide treatment group was obviously narrowed(P < 0.05),The volume of cerebral infarction in 14 d in each group was significantly smaller than 7d(P< 0.05).Conclusion:This research shows that:1.Danhong injection can promote the expression of VEGF after cerebralinfarction and angiogenesis,reducing infarct volume.2.The mechanism of Danhong injection to improve collateral circulation is probably by increasing the expression of VEGF in ischemic brain tissue and increasing blood vessel density in ischemic penumbra and improving blood supply,thereby reducing infarct volume.
