Objective: The aim of the study was to investigate the effect of Edaravone combined Danhong injection on the expression of MST1 in Hippo pathway in rats with acute ischemic stroke.Methods:A left middle cerebral artery occlusion(MCAO)model was established,SD rats were randomly divided into 5 groups with 6 rats in each group: sham operation group,model group,edaravone group,Danhong injection group,and edaravone combined with Danhong injection group(combined drug group),each group was divided into four subphase groups of 1,3,5,7 days.After corresponding drug treatment,neurological deficit scores were used to evaluate the neurological injury in rats.The expression of MST1 was detected by immunohistochemistry and q PCR,and its changes were observed.Result:1.Neurological deficit scores: The neurological deficit scores of the model group and each drug group at each time point were higher than those of the sham operation group(P<0.01);compared with the model group,the Edaravone group and Danhong injection group decreased significantly on the 5th and 7th days(P<0.01)after cerebral ischemia,while the scores of neurological deficits at the various time points after cerebral ischemia in the combination group were lower than the model group(P<0.01);and the Edaravone group compared with the Danhong injection group,the scores of neurological deficits on the 3rd,5th,and 7th days after cerebral ischemia in the combination group decreased(P<0.01).2.Immunohistochemical detection: Compared with the sham group,MST1 protein expression was increased in the model group and Danhong injection group at the same time point(P<0.01).Compared with the model group,the expression of MST1 protein was significantly decreased in the edaravone group on the 7th day after cerebral ischemia(P<0.01);there was no significant difference in the Danhong injection group(P>0.05);The expression of MST1 protein was significantly decreased in the last 4 phases(P<0.01).Compared with Edaravone group,MST1 protein expression was significantly increased in the Danhong injection group on the 7th day after cerebral ischemia(P<0.01);MST1 protein expression was decreased on the 5th and 7th days after cerebral ischemia in the combined group(P<0.01).Compared with the Danhong injection group,MST1 expression was significantly decreased at the same time point after cerebral ischemia in the combination group(P<0.01).3.q PCR detection:Compared with the sham-operated group,MST1 m RNA expression was increased in the model group and Danhong injection group at the same time point(P<0.01).Compared with the model group,the expression of MST1 m RNA was significantly decreased in the Edaravone group and the combination group on the 5th and 7th days after cerebral ischemia,and the combination group began to decline on the 3rd day(P<0.01);Compared with the Edaravone group,the expression of MST1 m RNA in the Danhong injection group was significantly increased at 5th and 7th day after cerebral ischemia(P<0.01),and the MST1 m RNA expression was decreased at the 7th day after cerebral ischemia in the combined group(P<0.01).Compared with Danhong injection group,the expression of MST1 was significantly decreased on the 1st,5th and 7th days after cerebral ischemia in the combination group(P<0.01).Conclusion:1.Edaravone play an anti-cerebral ischemia effect by inhibiting the expression of MST1,and Danhong injection has no obvious inhibitory effect on MST1.2.The effect of Edaravone combined with Danhong injection on the inhibition of MST1 was better than that of edaravone single drug,which could better play a role in brain protection.
