| 0bjective:Observation of zheng 1 new injury medicine for prevention and control of disease in the rat model at the end of the Achilles tendon effect,and the end of the organization of interleukin 1(IL 1),capillary epidermal growth factor(vascular endothelial growth factor,VEGF)and hypoxia inducing factor(HIF-1 alpha)expression,discusses the cheng 1 new injury medicine in the treatment of Achilles tendon terminal disease of HIF/regulatory mechanism of VEGF pathway.Methods:72 male Wistar rats were randomly divided into K group(blank control group),group T(pure jumps group)and group F(lo jump group),each group then based time points were randomly divided into group 4 weeks,6 weeks group,8 weeks group,each group of eight.K group natural feeding,T,F group adopts the method of "electric shock jump" mode,and group F from 4 weeks to start external treatment of zheng 1 new drug intervention.Observe the general situation of rats(hair,expression,physical condition,etc.)and after the jump and respectively in the end of 4,6,8 weekend take Achilles tendon tissue and HE staining to observe histomorphology changes,ELISA method to detect IL-1,VEGF,Western Blot method to detect alpha HIF-1 protein expression.Result:1.Gross observation: K group rats generally in good condition,build module(T,F group)in rats after the jump performance for look tired,fur messy,physical strength drops,but group is better than that of group T F.2.Effective hopping frequency: build module(T,F group)in the process of jumping downward trend on the whole,better than the group T and group F,there are significant differences in 4 ~ 7 weeks.3.HE staining tissue morphological observation: 4,6,8 weekend K group rat tendon fibers are arranged corrugated,"line" is clear,no inflammatory cell infiltration and new blood vessels.Weekend,4 T group rat tendon fiber is disorder,part of the fracture,the group F tendon fibre owe neat,can be found in a small amount of inflammatory cells infiltration.Weekend T 6 groups of tendon fibers were "glass",did not see performance at high tide and low tide,a small amount of inflammatory cells infiltration and new blood vessels.Group F tendon tissue more inflammatory cells infiltration and new blood vessels,faint.Weekend T group 8 tendon fibers were typical change "" glass samples,the tide line is " flame " and " high tide ",see more newborn blood capillary.cause group F,faint.4.Zheng 1 new injury control rats by pathological changes in protein expression at the end of the Achilles tendon changes:(1)the IL-1 in 4,6,8 weekend expression in T group were significantly higher than that of K and F group,and group F K group there was no significant difference.(2)of VEGF in the expression of T group is significantly higher than 4 weekends K group,group F with K group there was no significant difference.6,8 weekends compared between three groups had no obvious difference,but jumping group express a relatively high level.Alpha 4 weekends(3)HIF-1 the expression of T group rats is significantly higher than K group,there was no significant difference with the F group,6 weekend there were no significant differences,three groups of contrast in the 8th weekend T group was significantly higher than that of K and F group.conclusion:1.External treatment of zheng a new injury medicine rat Achilles tendon disease prevention and cure function,at the end to end disease pathological changes process is slow.2.Jump group of new blood vessels ingrowth Achilles tendon tissue in rat with HIF/VEGF signaling pathway on vascular endothelial cell growth factor regulation.3.Zheng 1 new drugs could inhibit the early inflammatory response,lower IL-1,HIF-1 alpha,at the same time increase the expression of VEGF,promote capillary.cause terminal area,improve local microcirculation status,in order to delay the process of the Achilles tendon terminal illness. |
