The Prevention And Treatment Effect Of Zheng's No. 1 New Wound Medicine On Rat Achilles Tendinopathy And The Mechanism Of Caspase12 Protein-induced Cell Apoptosis

Objective: To observe the effect of Zheng's No.1 new wound medicine on the control of the rat Achilles tendon terminal disease induced by electric shock and the mechanism of apoptosis induced by Caspase12 protein,and to explore the effect and mechanism of the external application of traditional Chinese medicine on the end of the Achilles tendon in rat induced model,and provide a theoretical basis for the clinical use of the drug.Methods: 72 male Wistar rats were randomly divided into group K(blank control group),group T(single jump group)and group F(dressing Jump Group).Each group was randomly divided into 4 weeks group,6 week group and 8 week group,each group was 8.Group K was fed naturally,group T and group F were modeled by electric shock jumping method,and group F was treated with Zheng No.1 wound medicine from fourth weeks.After observing the general situation of rats(hair,spirit,physical condition,etc.)and the number of effective jumping times,the end of the Achilles tendon was taken at the end of the fourth,sixth,eighth week respectively.The histomorphological changes were observed by HE staining,the apoptotic index was detected by Tunel method,and the Caspase12 protein was detected by Western Blot method.Results: 1.General situation observation: the rats in group K were generally in good condition.The rats in the model group(group T,group F)showed burnout,the fur was messy and the physical strength decreased,but the F group was better than the T group.2.The number of effective jumps: the whole group(group T and group F)showed a downward trend in the whole jump process,and the F group was better than the T group,and there was significant difference in 4~7 weeks.3.HE staining: at the end of fourth,sixth,eighth weeks,the tendon fibers in group K were all waved,"clear tide line" did not change the number of cells and the growth of neovascularization.At the end of fourth week,the tendon fibers of T group were partially broken,and the tendon fibers of group F were not regular.At the end of the sixth week,the rupture of tendon fiber in group T increased,showing "glass like change".There was no rise in tide and ebb tide.There was a significant increase in the number of cells and a small amount of neovascularization.Group F showed obvious cell proliferation and neovascularization,and the tidal line was blurred.At the end of the eighth week,the tendon tissue in group T showed a typical "glassy change".The tide line showed "flame like change" and "high tide".The number of cells increased.In group F,more new capillaries were seen and the tide line was blurred.4.Cell apoptosis detection:(1)Intra group analysis: group K and group F showed a downward trend from fourth weeks to eighth weeks,while group T showed a slow upward trend.There was no significant difference between group K and group T,P > 0.05;there was significant difference between group F and group fourth and eighth weeks,P < 0.05.(2)Group analysis: fourth weeks,group T and group F were significantly higher than group K,and there was significant difference between group K,F and T group,P < 0.05;sixth weeks,T group was higher,T group and K group were significantly different,P < 0.05;eighth weeks,T group was significantly higher than the two groups,and there were significant differences between the two groups.The expression of 5.Caspase12 protein:(1)In group K and group F,the changes in group F and group T were first and then descended,and there was no significant difference between group K,T and F in the comparison and analysis in group,P > 0.05.(2)Group results: fourth weeks,group T and group F were higher than group K,and there was significant difference between group K and T group and F group,P < 0.05,T group and F group had no significant difference,P > 0.05.At sixth weeks,T group was higher than the other two groups,and there was significant difference between K group,group and group.Eighth weeks,the group was higher than the other two groups.There was a significant difference between the K group and the T jumping group,P < 0.05.There was no significant difference between group T and group F,and P > 0.05.Conclusion: 1.Zheng's No.1 new wound medicine can effectively delay the pathological changes of terminal disease and prevent and treat Achilles tendon terminal disease in rats.2.during the formation of the terminal disease,it was found that the expression of Caspase12 protein was associated with apoptosis,and Caspase12 protein could effectively regulate the development of cell apoptosis through the endoplasmic reticulum pathway.3.Zheng's No.1 new wound medicine can effectively inhibit the apoptosis of tissue cells by regulating the expression of Caspase12 protein,which can delay the pathological changes of the terminal and prevent the Achilles tendon terminal disease.
