Objective:To study the protein expression of type ? collagen fiber(Col ?),type III collagen fiber(Col III),aggrecanase-1(ADAMTS4)and aggrecanase-2(ADAMTS5)in the development of Achilles tendinopathy in rats.Methods:Ninety-six male SD rats aged 8 weeks were randomly divided into blank group(C group),simple jump group(J group),simple compress group(M group),and jump compress group(JM group).They were divided into 4,6,and 8 week groups according to different sampling times.Group C was fed naturally without intervention;Group M began to receive external application of Zheng's No.1 new wound medicine at the 4th week;Group J and Group JM were modeled by "electric shock jumping method";Group JM was given external application of Zheng's No.1 new wound medicine at the 4th week.MAIN OUTCOME MEASURES: The general condition of rats and the number of effective jumps,the morphology of tendon fibers were observed by HE staining,and the protein expression of ADAMTS4,ADAMTS5,Col I and Col III in rat Achilles tendon was determined by Western Blot.The data were analyzed by independent sample T test and two-way ANOVA.Results: 1.Number of effective jumps: Both group J and group JM showed a decreasing trend.At week 4,group J was slightly different from group JM,but no significant difference was observed.From week 6 to week 8,the number of effective jumps in group J was significantly less than that in group JM,and there was a significant difference.2.Morphological results of HE staining: At the 4th week,the collagen fibers of group J showed a tendency of destruction,and some collagen fibers were broken.No significant fragmentation of collagen fibers was observed in the JM group.At the 6th week,there were partial breaks in collagen fibers and pathological changes of tendinopathy with increased nuclei in group J.In the JM group,there were a few breaks in the tendon fibers and the arrangement was less regular.At the 8th week,the collagen fibers in group J were fractured in large area,and new blood vessels grew into the fibrocartilage area.In the JM group,the collagen fibers were disordered,with a few tendon fibers broken and a small amount of inflammatory cell infiltration.3.Expression of Col I: There was no significant difference in the data between the 4th week and the 6th week,only the trend changed.At week 8,the downregulation in group J was significantly different from that in group C;there was a highly significant difference compared with groups M and JM.The highest up-regulation was observed in the JM group,with a highly significant difference compared with the J group.4.Col III expression: At week 6,the up-regulation in group J was significantly different from both groups C and M;the up-regulation in group JM was significantly different from both groups C.At the 8th week,the up-regulated expression was the highest in group J,which was significantly different from both groups C and M;the slightly down-regulated expression in group JM was significantly different from that in group C.5.ADAMTS4 expression: There was no significant difference in the data between week 4 and week 6,only the change of trend.At week 8,the up-regulation in group J was significantly different from the other three groups.6.ADAMTS5 expression: there was a highly significant difference between the sixth week and the eighth week in group J;group J showed a trend of first decreasing and then slowly increasing.There was a significant difference between the 4th and 8th week in the JM group;the JM group showed a continuous upward trend.At week 6,down-regulation in group J was significantly different from both groups C and M,and highly significantly different from JM;there was no significant difference between group JM and groups C and M.At week 8,up-regulation in JM group was significantly different from C,M and J groups.The results showed that Zheng's No.1 new wound medicine had certain preventive and therapeutic effects on tendinopathy in rats.2.2.From the results of protein expression,it is known that Zheng's No.1 new wound drug may have a preventive and therapeutic effect on tendinopathy in rats by down-regulating the expression of ADAMTS4 and up-regulating the expression of ADAMTS5.
