Objective:Observe the effect of electroacupuncture at neck clip ridge point of CS model rabbit to Chondrocyte apoptosis rate and the IOD of JAK2 and STAT3.Explore the possible mechanisms of the treatment to provide experimental basis for clinical acupuncture in the treatment of CS.Methods:According to the random number table method,18 rabbits were divided into blank group,model group and electroacupuncture group,6 rabbits in each group.Model group and electroacupuncture group were made by the method of flexed neck posture for long-term.After the success of the CS models,the electroacupuncture group was treated with electroacupuncture for 21 days.After the end of the treatment,the animals were killed by injection of air embolism in the auricular vein.The tissue of C4-7 intervertebral disc was taken and the cartilage endplate was separated.The morphological structure of chondrocytes was observed,the percentage of TUNEL positive cells was detected and the IOD of JAK2 and STAT3 was detected under light microscope.Results:1.The cartilage cell morphology observation: There were obvious apoptotic changes in the model group than that in the blank group,and some apoptotic changes were found in the electroacupuncture group compared with the blank group.2.Positive rate of apoptotic chondrocytes: the model group and the electroacupuncture group was higher than the blank group(P<0.01),the electroacupuncture group was lower than the model group(P<0.01).3.The IOD of JAK2 and STAT3: the IOD of JAK2 in the model group and the electroacupuncture group was lower than that in the blank group(P<0.01),the IOD of JAK2 in the electroacupuncture group was higher than that in the model group(P<0.01).The IOD of STAT3 in the model group was lower than that in the blank group(P<0.01),the IOD of STAT3 in the electroacupuncture group was lower than that in the blank group(P<0.05),the IOD of STAT3 in the electroacupuncture group was higher than that in the model group(P<0.01).Results:1.Electroacupuncture at the neck clip ridge point can promote the structural repair of cervical vertebral disc cartilage cells.2.Electroacupuncture at theneck clip ridge point can regulate JAK2 and STAT3,reduce the apoptosis of cervical intervertebral disc cartilage,delay the degeneration of intervertebral disc,and regulate cervical spondylosis.
