Objective:The purpose of this study was to investigate the mechanism of electroacupuncture on the expression of JAKT2/STAT3 m RNA in rabbit intervertebral disc chondrocytes by electroacupuncture at Jiaji point in order to explore the mechanism of electroacupuncture in the treatment of cervical spondylosis.Methods:Eighteen purebred New Zealand rabbits(half male and half female)were randomly divided into group A(model group C).After the model was successful,group A and B were not treated.Group C was treated with electroacupuncture.The time of treatment was 21 days.After the treatment,all animals were killed and C3 C7 intervertebral disc was taken out.Cartilage endplate was separated and morphological structure of cartilage cells was observed.JAK2 and STAT3 m RNA expression was detected by RT PCR method.Results: 1.Compared with the control group,the model group had lower mental function,less movement,less diet,stiff neck muscles and less flexible neck movement.2.Observation of chondrocyte morphology and structure: there were obvious apoptosis changes in the model group than in the blank group.Fluorescence quantitative PCR was used to detect the relative expression of JAK2 / STAT3 m RNA in rabbit intervertebral disc chondrocytes: the relative expression of JAK2 m RNA in EA group was lower than that in control group(P < 0.05),and the relative content of JAK2 STATm RNA in EA group was significantly higher than that in model group(P < 0.01).Expression: compared with the blank group,the relative content of the electroacupuncture group was decreased(P < 0.05),and the relative content of the electroacupuncture group was higher than that of the model group(P < 0.05).Conclusion: 1.Electroacupuncture at Jiaji point can inhibit the apoptosis of chondrocytes ofintervertebral disc and promote the self-repair of chondrocytes of intervertebral disc.2.Electroacupuncture at Jiaji point could increase the expression of JAK2 + STAT3 m RNA in the chondrocytes of intervertebral disc,suggesting that electroacupuncture could inhibit the apoptosis of chondrocytes and delay the degeneration of intervertebral disc.
