ObjectivePreliminary study on the effect of abdominal massage on antidepressant-like behavior and iris in CUMS rats MethodsPart 1,Systematic review and Meta analysis of massage in the treatment of depression.Through the computer search of Pub Med,China knowledge Network(CNKI),Wanfang database,VIP(VIP)and Chinese biomedical literature(CBM)database,the domestic and foreign RCT literatures of massage in the treatment of depression from March to March 2020 were searched,with Hamilton Depression scale(HAMD),Baker Depression scale((BDI))and self-rating Depression scale(SDS)as outcome indicators.Revman5.3 software was used for literature quality evaluation and Meta analysis,heterogeneity test and funnel chart observation.Part 2:To establish the antidepressant effect platform of rubbing abdomen.In this study,chronic unpredictable mild stress(chronic unpredictable mild stress,CUMS)combined with single cage feeding was used to establish a rat depression model.The sugar water preference test(Sucrose preference test,SPT),forced swimming test(Forced Swin Test,FST)and the change of body weight were used as the indexes of depression-like behavior of rats.According to the index of sugar preference and body weight,the patients were randomly divided into blank group,model group and abdominal massage group.the stress methods included electric shock,restraint,ice water swimming,tilted rat cage,tail clamping,noise,wet cushion,overnight lighting,water prohibition and fasting.Two of them were randomly selected every day to give the same stress to the model group and the abdominal massage group,and the abdominal massage group received 10-minute abdominal massage intervention 1 hour before modeling,and after 3 weeks,the abdominal massage group was given 10 minutes of abdominal massage intervention 1 hour before modeling,and after 3 weeks of modeling,two of them were randomly selected to give the same stress to the model group and the abdominal massage group.Then the changes of SPT,FST and body weight of rats in each group were measured.Part 3:The molecular biological method was used to explore the antidepressant mechanism of rubbing abdomen.On the basis of successful establishment of the model,(enzyme linked immunosorbent assay,ELISA)was determined by enzyme-linked immunosorbent assay(Elisa),Western blotting(Westen Blot,WB)and gene amplification fluorescence detection(Real-time Quantitative PCR,RT-q PCR)techniques were used to detect Iris protein and gene expression in serum,hippocampus and cortex,respectively.ResultsA total of 1094 articles were searched and 16 articles were included in this study.(HAMD)score of Hamilton Depression scale: the results of Meta analysis showed that massage treatment could significantly reduce the HAMD score of patients [SMD=-1.72,95%CI(-2.31)].Simple massage is better than oral western medicine group in HAMD score,massage + routine nursing + emotional nursing is better than routine nursing + emotional nursing in HAMD score.SDS score [MD=-8.51,95%CI(-11.93 mai Murray 5.10)].To establish an animal experimental platform for rubbing abdomen and antidepressant.The results showed that there was no significant difference in sugar preference and body weight among blank group,model group and abdominal massage group before modeling(P > 0.05).(1)appearance and general state: the coat color of rats in the blank group was normal,lively and active,responsive and in good condition.After chronic stress,the rats in the model group showed increased defecation and urination,reduced activity,irregular hair,yellow color and slow mental state.The abdominal massage group was in a good state of mind,neat hair and curious about new things.(2)Sugar preference: after modeling,the sugar preference in the model group was significantly lower than that in the blank group(P < 0.01),and there was significant difference between the abdominal massage group and the model group(P < 0.05).(3)forced swimming: after modeling,the immobility time in the model group was significantly higher than that in the blank group(P < 0.01),and the immobility time in the abdominal massage group was significantly lower than that in the model group(P < 0.05).(4)weight change: there was no significant difference in body weight before modeling,but there was significant difference after intervention(P < 0.01).The results of Elisa detection showed that the content of serum Iris in the model group was lower than that in the blank group,the expression of serum Iris protein in the model group was higher than that in the model group,the expression of Iris protein in the hippocampus in the model group was lower than that in the blank group,and the expression of Iris protein in the hippocampus in the model group was higher than that in the model group.There was no significant difference in the expression of iris in the cortex among the blank group,the model group and the rubbing abdomen group(P > 0.05).Conclusion 1.Meta analysis suggested that massage was effective in the treatment of depression.two?.Abdominal massage intervention can improve the depression-like behavior of CUMS depression model rats and has antidepressant effect.3.Abdominal massage increased the expression of Iris protein in serum and hippocampus of CUMS rats.
