Objective: In this study,Ureaplasmaurealyticum(Uu)was used to constructed NGV mouse models.In order to clarify the changes in the vaginal microecology of the NGV mice treated by Bixie Fenqing Decoction,and study the changes in serum Changes in immune factors and pathological changes in genitals evaluate the efficacy.It will reveal the mechanism of efficacy,and provide a theoretical basis for the wider promotion of traditional Chinese medicine treatment of NGV.Methods: 1.SPF female BALB/c mice were divided into 7 groups: the normal control group,the model group,the positive Chinese medicine Jingang Teng syrup group,and the positive western medicine Roxithromycin group and Bixie Fenqing Decoction high dose group,Bixie Fenqing Decoction middle dose group,Bixie Fenqing Decoction low dose group,10 mice in each group.Except the normal group,mice in the other groups were pre-treated with estrogen and infected with the Uu.2.All groups on the 7th,14 th,and 21 st days were detected the mental state and weight.Vaginal mucosal secretions are collected and Gram stained to evaluate the vaginal microecological restoration of mice in each administration group.3.On the 7th,14 th,and 21 th days of administration,the serum of each group of mice was collected,CD3~+,CD4~+,and CD8~+ changes in serum T cell population were detected using(flow cytometer,FCM)to clear the effect of Bixie Fenqing Decoction on the body's immune regulation.4.Mice were sacrificed after the 21 st day of administration,and genital tract mucosa samples and serum were collected from each for HE staining and IL-6 immunohistochemical staining to analyze the effect of Bixie Fenqing Decoction on the inflammatory response of Uu induced NGV model.To clarify the antagonistic effects of Bixie Fenqing Decoction on NGV pathological damage caused by Uu,the levels of pro-inflammatory cytokines(TNF-?,IL-1? and IL-8)and anti-inflammatory cytokines(IL-4 and IL-10)were detected by ELISA.Results: 1.Bixie Fenqing Decoction improved behavioral indicators such as mental state and weight loss of NGV model mice caused by or Uu.On the 21 st day of administration,Jingang Teng syrup group,the mid-and high-dose groups of Bixie Fenqing Decoction had better improvement effects than the western medicine roxithromycin group,high-dose Bixie Fenqing Decoction showed optimal improvement.2.Gram staining showed that on the 7th day of administration,the roxithromycin group had the best inhibitory effect on the number of Gram-positive bacteria in the vaginal mucosa of NGV mice(P<0.01).However,with the prolonged administration time,the inhibitory effects of Jingang Teng syrup group and Bixie Fenqing Decoction on the gram-positive bacteria in vaginal mucosa of NGV mice were better than that of roxithromycin group(P<0.01).It suggested that Qifenqing Decoction could significantly inhibit the bacterial infection in the vaginal area.3.Compared with the control group,the levels of CD3~+,CD4~+,and CD4~+/CD8~+ in the serum T cell population of the model group were decreased,and the level of CD8~+ was increased.After the 7th,14 th,and 21 st days of Bixie Fenqing Decoction administration,compared with the model group,the levels of CD3~+,CD4~+,and CD4~+/CD8~+ in the serum T cell population gradually increased,and the CD8~+ level decreased.In addition,the effect of improving the levels of CD3~+,CD4~+ and CD4~+/CD8~+ on the Jingang Teng syrup group,mid-and high-dose groups of Bixie Fenqing Decoction was significantly better than that of roxithromycin group,suggesting that the Bixie Fenqing Decoction had good effects of immunity on NGV mice,as the dosage of Bixie Fenqing Decoction increased,the improvement effect became more obvious.4.HE staining results showed that Bixie Fenqing Decoction displayed a good repaireffect on vaginal mucosa of Uu induced NGV mice.Compared with roxithromycin group,mid-and high-dose groups of Bixie Fenqing Decoction had more significantly inhibitory effect on inflammatory infiltration of the vaginal wall tissue.IHC results showed that the expression level of IL-6 in Bixie Fenqing Decoction high dose group was lowest compared with the model group.The ELISA results showed that,compared with the model group,the proinflammatory factors TNF-?,IL-1? and IL-8 were inhibited after Bixie Fenqing Decoction treatment(P<0.01),and the anti-inflammatory factors IL-4 and IL-10 have were elevated in Bixie Fenqing Decoction group(P<0.05).Moreover,there is a certain difference between low,medium and high doses of Bixie Fenqing Decoction.The dose-response relationship suggested that Bixie Fenqing Decoction had a better anti-inflammatory effect than roxithromycin in the Uu induced NGV model.Conclusion: Bixie Fenqing Decoction could improve the general condition and weight of Uu induced NGV mice,and significantly reduce the number of gram-positive bacteria in vaginal area of Uu induced NGV mice.Compared with the inflammatory factors and immunological indicators of mice in each group,the results showed that Bixie Fenqing Decoction improved the changes of Uu induced NGV mice type immune indicators and inflammation indicators.The treatment effect of NGV mice is better than that of roxithromycin groupand high-dose Bixie Fenqing Decoction shows the best treatment effect,suggesting that Bixie Fenqing Decoction had a better therapeutic effect on non-gonococcal vaginitis caused by Uu compared with the current clinical antibiotic therapy.It provided theoretical basis for Chinese medicine therapy to improve vaginal micro-ecology,improve body immunity to treat non-gonococcal vaginitis caused by Uu.
