| Objective:To observe the role of Bixie Chubi Decoction in regulating the inflammatory factors of kidney tissues of rats,and to explore the mechanism of the treatment of uric acid nephropathy.Method:The experiment was divided into two parts.The first part was the animal experiment section(vivo experiment).The experimental animals were randomly divided into 6 groups(blank group,model group,western medicine group,low dose group of traditional Chinese medicine,middle dose group,high dose group),then made the preparation of uric acid nephropathy rat model(adenine+ethambutol),modeling and administration were done at the same time.The general situation of rats was observed.The rats were killed and the preparation of rat kidney pathological sections was made to observe the pathological status of renal tissue in each group.The serum uric acid,creatinine,urea nitrogen of rats were detected,the protein production status of NF-?B was detected by the method of immunogenetic,the expression of each inflammatory indicator in rat kidney was detected by the method of Western blot,the mRNA expression of each indicator in rat kidney was detected by the method of RT-PCR.The second part of the experiment was the cell experiment section(vitro experiment).The cell cultivation and cell passage of rat macrophage NR83 83 cells were made;each group of cells was stimulated by LPS solution,and the preparation of serum with different concentrations of Bixie Chubi Decoction.The protein expression and mRNA expression of macrophages in rats were detected by the method of Western blot and RT-PCR,and the differences between each group were observed.Result:1.Results of Animal experimental study:the effect of modeling method of adenine and ethambutol was obvious,and the pathological status of uric acid nephropathy in model group was significant.The renal pathological results of rats indicated that the decoction could reduce the deposition of uric acid effectively and release the infiltration of inflammatory cells and dilatation of renal tubules.Compared with the model group,the uric acid,creatinine and urea nitrogen of the rats in other groups was significantly decreased(P<0.01);the generation of NF-?B protein and the protein expression and gene expression of each indicator in the kidney issues were decreased(P<0.05),and the comparison between the medium dose group and the high dose group was more obvious(P<0.01).2.Results of cell test:Bixie Chubi Decoction could regulate the high expression of each indicator of protein and gene of NR8383 cells in rats(P<0.05);among all the statics,the result of the highest concentration is the most obvious(P<0.01).Conclusion:The experiment demonstrates that Bixie Chubi Decoction can reduce the expression of inflammatory factors in rats with urinary acid nephropathy,reduce blood uric acid,relieve renal injury,and the result can be confirmed by vitro experiments. |
