ObjectiveTo clarify the expression levels of nuclear related antigen Ki67 and Epidermal growth factor receptor(EGFR)in adenocarcinoma of esophagogasetric(AEG)tissues,and to explore the expression levels of Ki67 and EGFR and their clinical pathology The correlation between parameters and their impact on patient survival and prognosis.With a view to providing new targets and new ideas for the treatment of AEG.MethodsAdenocarcinoma tissue samples from 165 patients with esophagogastric tumors from 2007 to 2015 were collected.Immunohistochemistry(IHC)method was used to detect the expression of Ki67 and EGFR in AEG patients.At the same time,the clinical characteristics and pathological data of patients were collected,including age,gender,histological type,tumor size,degree of differentiation,TNM stage(T represents the depth of primary tumor invasion,N represents the regional lymph node metastasis of the patient,and M represents distant Metastasis),surgical methods,etc.were retrospectively analyzed.SPSS22.0 statistical software was used to analyze the data.The paired t test was used to compare the expression levels of Ki67 and EGFR in tumor tissues.The?2 test was used to analyze the correlation between Ki67 and EGFR high expression level group and low expression level group in clinicopathological parameters.Multivariate factors were used.COX regression analysis was used to analyze the correlation between multiple clinicopathological factors and survival status.The Kaplan-Meier method was used to draw the survival curves of the overall survival rate of Ki67and EGFR high expression level group and low expression level group,and Log-rank test was used to compare the differences.Survival differences between groups.The difference was statistically significant at p<0.05.Results(1)The overexpression rate of Ki67 protein in primary AEG was 54.5%,which was related to the tumor diameter(?~2=9.974,p=0.002)and the number of lymph node metastases(?~2=4.727,p=0.030).Postoperative recurrence and metastasis(?~2=5.509,p=0.019)and adjuvant chemotherapy(?~2=6.059,p=0.014)were related.It has nothing to do with the patient's gender,age,surgical method,Siewert classification,WHO histological classification,neurovascular invasion,vascular invasion,and clinical stage of TNM(p>0.05).The overall survival rate of the high expression group was lower than that of the low expression group.(2)The overexpression rate of EGFR in AEG was 46.7%,which was related to tumor diameter(?~2=13.74,p<0.001),TNM pathological stage(?2=8.371,p=0.004),and postoperative recurrence and metastasis(?2=15.884,p=0.0006).It has nothing to do with the patient's gender,age,surgical method,Siewert classification,WHO histological classification,vascular and vascular invasion,surgical complications,lymph node metastasis and postoperative adjuvant chemotherapy(p>0.05).The overall survival rate of the high expression group was lower than that of the low expression group.(3)The results of univariate analysis revealed significant differences between tumor differentiation,Ki67 expression,and EGFR expression(p<0.05);further from the multivariate COX regression analysis,the degree of tumor differentiation(B:0.912,OR:0.402,OR95%CI:0.199-0.810,p:0.011)and Ki67 expression(B:0.860,OR:2.364,OR 95%CI:1.145-4.881,p:0.020)are independent risks that affect the prognosis of AEG Factors(p<0.05).Conclusion Ki67 and EGFR are up-regulated in adenocarcinoma of the esophagogastric junction,and Ki67 and tumor differentiation are independent factors affecting the survival prognosis of AEG,which provides new ideas for the development of AEG targets.
