Buddleja officinalis Maxim which is a kind of traditional medicine with the activities of anti-inflammatory,antioxidant,bactericidal and immunomodulatory functions is the dry flower bud and inflorescence of the genus Buddleja L in the Loganiaceae family.There had been several focuses on Buddleja officinalis Maxim all over the world so far.The compounds of Buddleja officinalis Maxim are mainly composed of flavonoids,terpenes,phenylethanolic glycosides and volatile oil based on this study.Buddleja officinalis Maxim had been found that it has the pharmacological activities of antibacterial,antioxidant,hypoglycemic,improve immunity,anti-ocular diseases pharmacological activities,and liver protection has not been reported.In this work,the active ingredients were extracted with 75%ethanol and ethyl acetate,successively.Then,ethyl acetate extracts were obtained.The monomeric compounds in Buddleja officinalis Maxim were obtained by the silica gel,reversed phase silica gel and Sephadex LH-2 column.Then,structures of those were identified by Mass Spectrometry(MS),Nuclear Magnetic Resonance(1H-NMR,13C-NMR).There were 32 compounds isolated from the Buddleja officinalis Maxim,i-ncluding 10 pentacyclic triterpenes(?-amyrin,cycloeucalenone,lupeol acetate,?-amyrenone,24-oxo-31-norcycloartanone,cycloeucalenol,lupeol,ursolic acid,s-tigmasterol,?-spinasterol),8 flavonoids(acacetin,apigenin,linarin,luteolin,rutin,luteolin-7-O-glucoside,cosmosiin,kaempferol-3-O-rutinoside),6 organic a-cid lipid compounds(laurin,methyl palmitate,methyl linolenate,1,2-O-isopro-pylidene-3-O-palmitoylglycerine,glyceryl monooleate,1,3-tetradecocic acid glyc-etine two ester),2 phenylethanoid glycosides(verbascoside,cistanoside F),2 alcohol compounds(cis-11-hexadecene-1-ol,1-octanol),1 aromatic compou-nd(ethylparaben),1 iridoid compounds(methylcatalpol),1 heterocyclic co-mpound((S)-2-(2'-Hydroxyphenyl)-4-hydroxymethyl-4,5-dihydrothiazole)and 1 saccharide compound(diacylgalactolipid ?).There are 5 pentacyclic triterpenes,4 organic acid lipid compounds,2 alcohol compounds,1 flavonoid,1 steroid,1 iridoid,1 aromatic and 1 saccharide among the 32 compounds isolated from Buddleja officinalis Maxim.Moreover,the above 16 compounds were firstly obtained from Buddleja officinalis Maxim.In this work,different eluent segments of the ethyl acetate extract of Buddleja officinalis Maxim were taken to study its effect on normal liver cells.The results showed that most extracts of Buddleja officinalis Maxim had no damage effect on normal liver cells,providing a basis for the separation and purification of the liver-protecting components of Buddleja officinalis Maxim and further application research.
