Zanthoxylum bungeanum Maxim belonged to Rutaceae was a traditional medicinal and edible material in China. Alkaloids have anti-inflammatory antipruritic, analgesic and anti-tumor sterilization and other physiological activity. Zanthoxylum bungeanum Maxim is rich in alkaloids, although the research on that was less at home and abroad.The determination, purification, separation, sensory evaluation and pharmacology activity of alkaloids were studied in the paper. The main experiment content was as follows:Acid-base titration was used to determinaed the assaying of alkaloids. Orthogonal test was adopted to optimize the extracting process of alkaloids from Zanthoxylum bungeanum Maxim. Four factors of the extraction temperature, time, ethanol concentration and pH were studied. The results showed that the factor on the extraction rate were in a sequence:ethanol concentration>pH> extraction temperature >extraction time. The optimum extracting was as follows:70% alcohol as extraction solvent, pH 3, extraction time conditions 3h, and extraction temperature 60℃.The yield of total alkaloids was 0.8898% under these conditions.The purification with macroporous resin of alkaloids from Zanthoxylum bungeanum Maxim was studied. The NKA-9 macroporous resin was used to purify total alkaloids. Thin layer chromatography and ultraviolet-spectrophotometer assay were used to identify Zanthoxylum alkaloids. The results showed the optimum conditions were that:2h adsorption,3BV of water,50%,70% ethanol for elution. The 70% ethanol elution fractions were alkaloids, and 50% ethanol elution fraction was flavonoids. Under these conditions, the alkaloids rom Zanthoxylum bungeanum Maxim was separated.High-speed counter-current chromatography (HSCCC) with a two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water (5:5:7:7, v/v) was applied to the isolation and purification of alkaloids from Zanthoxylum bungeanum Maxim. The lower phase was used as the mobile phase and performed at a flow rate of 2mL/min, while the apparatus rotated at 800r/min, and detected at 254 nm. Two kinds of alkaloids were obtained and yielded 130mg (A),15mg (B) from 800mg of crude extract in a one-step separation. A and B were identified by Thin Layer chromatography (TLC), and the results showed that the two fractions were alkaloids, with the purity of 92.8% and 85.4%, as determined by high performance liquid chromatography (HPLC).Sensory evaluation was one subiect for the development, measurement, analysis and description of human beings through the sight, smell taste, touch, heariong and sense of feeling.At present, the mian direction of sensory science was developmented to the objective and semi-quantitative direction.This study was refered to the degree of determiniation of capsaicin. Flax taste components of the test samples were extracted by ethanol, and filtered then. The taste components were made from different numb-taste concentrations by sugar solution. The maxim was finded out by sensory analysis, and the numb-taste level was calculated through that. The results showed that the levels of ethanol extraction and macroporous elution fraction were seven, eight.Itchy skin disease was common, frequently-occurring disease, which caused by some media including histamine, proteases, vasodilator peptide hormone to stimulate the skin. Treatment for skin itching was made the improve core. Tt was better if anti-inflammatory, analgesic and inhibitory effect of purulent skin infection were considered.The pharmacological effects of Zanthoxylum alkaloids were discussed in this chapter. Filter paper method, mouse hot plate method, xylene-induced inflammation method and dexrran-40 itchy skin antibacterial were used to studied the antibacterial, analgesic, anti-inflammatory, antipruitic effect of Zanthoxylum alkaloids. The results showed that Zanthoxylum alkaloids were strong in antibacterial, analgesic, anti-inflammatory, antipruitic effect. Zanthoxylum alkaloids were worthy of further resear and development.
