Sanguisorbae Radix,a member of Rosaceae plants,is the dry root of Sanguisorba officinalis L.or Sanguisorba officinalis L.Var Longifolia(Bert.Yu et li).With a function of “cooling blood and hemostasis,detoxification and converging scores”,it has been widely used in the fight against surgical bleeding,burns,proctitis,ulcerative colitis as well as pyelonephritis in clinical.The main constituents include triterpenoid saponins,flavonoids,phenols,etc,among which the phenol category is quite active in the treatment for elevating white blood cell,anti-cancer,anti-oxidant and anti-bacterial.By tracking the literatures we find that there's limited research on the phenol constituent,besides,the current methods for phenol extraction and separation have a very low yield proportion.In order to investigate the phenol constituent in a deeper way and provide a scientific guidance for pharmacodynamics-material study,we report a optimization extraction and separation method of Sanguisorba officinalis and as well investigate the phenol constituent.The experiment is divided into two parts,described as follows:The first part of this work is about the optimization extraction and separation methods for phenols.We respective compare the extraction solvent,ethanol and acetone.The extraction method: macroporous resin adsorption and gel precipitation.The extraction packing: MCI,D101,Diaion HP-20 and Toyopearl HW-40;Elution system,positive and negative phase elution system.The results indicate the ultimately optimization extraction and separation method is: 70% ethanol extraction,followed by diacolation D101 macroporous resin to enrich phenols constituent,and then Diaion HP-20 and Toyopearl HW-40 are employed for further separation.The second part is about the identification and separation research of Sanguisorba officinalis L.chemical constituents.The methods mentioned in the first part is employed to separate and extract those chemical constituents,which are then purified with preparation chromatography,semipreparative chromatography and recrystallization methods.The identification procedure is then ran with the help of Spectra(1H-NMR and 13C-NMR)analytical method.We finally identified 20compounds(18 phenols,a Single post diglucoside and a Phenylethyl alcohol compounds),thus: gallic acid(1),protocatechuic acid(2),3,4-dihydroxybenzaldehyde(3),methyl gallate(4),ethyl gallate(5),3,4-dihydroxy-5-methoxybenzoic acid methyl ester(6),3,4-dihydroxy-5-methoxybenzoic acid ethyl ester(7),isolariciresinol 9'-?-D-xylopyranoside(8),(+)-piresil-4-O-?-D-glucopyraside(9),(+)-syringaresinol(10),1-(4-ethylphenyl)-1,2-ethanediol(11),ethyl-3,4-dihydroxy benzoate(12),3,4-dimethoxy-5-hydroxybenzoic acid methyl ester(13),4-(4-hydroxyphenyl)-2-butanone(14),miyaginin(15),geranyl-1-O-?-D-xylopyranosyl-(1-6)-?-D-glucopyranoside(16),prunin(17),confusoside(18),garbanzol(19),Icariside E4(20),among which compounds 3-20 are all first extracted in genus,among which compounds 2-20 are all first extracted in Sanguisorba officinalis L..
