In this paper, the extraction and separation techniques of acteoside from Buddlejaofficinalis Maxim and sprout of Sesame were studied. Optimizing the process by ResponseSurface Method and orthogonal experimental design. The quality level of acteoside fromBuddleja officinalis Maxim and sprout of Sesame was also studied. The main study contentand fruit is listed as follows.(1)Studing the separation techniques of Buddleja officinalis Maxim by orthogonalexperimental design. Separation conditon of Glycosides tapered column when optimizedwas15°chromatography column,silica gel: the quality of the sample ratio of75:1, elutionvelocity of18ml/min. Under the condition, the percent recovery of acteoside fromBuddleja officinalis Maxim was85.9%, the purity of acteoside could reach more85%.(2)Determining the content of acteoside from Buddleja officinalis Maxim and sprout ofSesame by HPLC(High Performance Liquid Chromatography), the results were: thecontent of acteoside from Buddleja officinalis Maxim was1.13%, RSD(Relative StandardDeviation) was0.89%, the convery was99.34%, RSD was0.96%, the content of acteosidefrom Sesame Seed bud was0.76%, RSD was0.88%, the convery was98.54%, RSD was0.94%.(3) Researching the sprout of Sesame production process by Response Surface Method.Validated experimen show that the optimization condition were: soaking time、 theconcentration of ethanol、 temperature were5h、8%、22℃,the germination index ofSesame Seed bud could reach24.82in theory.(4) Studing the separation techniques of sprout of Sesame by Response Surface Method.The experiments show that: when the concentration of ethanol was90%, the liquid ratiowas30:1, the extraction temperature was59℃, extraction time was240min, the percentrecovery of acteoside from sprout of Sesame was0.48%.(5) Studying the anti-Alzheimer’s disease of the extraction from sprout of Sesame onmice experiment. Experimental results showed, the extraction within0.1-0.4g/kg dosecould increase the activity of SOD, reduce the content of MDA.In this concentrationrange, the extraction could shorten the incubation period of the tested mice, increase times and extend the stay time in first quadrant.(6)Researching the compression technology of extractive of sprout of Sesame, theexperiment show that: choosing60%of ethonal as unguent, drying time was60min,Lactose: microcrystalline cellulose ratio is2.5:1, under the condition, test block, aspect,disintegration time limited and friability were fit the provision.
