| Magnolia officinalis cortex, asa geo-authentic crude drug of Chongqing China, isthe dry root bark and branches ofMagnolia officinalis Rehd. et Wils.or Magnoliaofficinalis Rehd. et Wils. var. biloba Rehd. et Wils. Mainly chemical ingredients containvolatile oil, lignans, alkaloids, polysaccharides and so on, which possess versatilepharmacologic activitiesincluding antibacterial, anti-inflammatory. At present, theresearch of Magnolia officinalis cortexmainlyfocus on its lignans components, magololand honokiol, extraction process and their effect of anti-inflammatory,the otherbioactivityingredients, however,were reported rarely. In this paper, lignans, alkaloids,polysacharides components were isolated fromMagnolia officinalis cortex, based on thepreparation of bioactivity ingredients, their antioxidant activity were evaluatedbychemiluminescence method.These results provided a scientific basis for the furtherdevelopment and utilization of Magnolia officinalis.What’s more, alow sampleconsumption and integrateddetection system were builted,which based on microfluidictechnology and the theory of chemiluminescence reaction. This microfluidicchemiluminescence detection system would besuggested to evaluate antioxidant activityof drugs or screeningthe antioxidants from natural products.The main researchdetails and results are as follows:1. To aim at theantioxidantingredients ofMagnolia officinalis. Differentextractswere extractedby water,60%ethanol and80%ethanol, and thecontentofmagolol,honokiol and total phenol were detected by HPLC andFolin-Ciocalteurespectivelyto help preliminary understand composition andpredict theantioxidant activity of different extracts. As a result, the80%ethanol extract containedthehighest content of magolol,honokiol and total phenol, respectively up to209.4mg/g,159.8mg/g and228.3GAE mg/g. In addition, three kinds of mainly potentialantioxidants include crude polysaccharides, total alkaloids and lignans were isolated bythe method of water extract-alcoholprecipitate, organic solvent extract and macroporousresin, respectively. As a result, the content of polysaccharides,alkaloids andlignans was50.8%,63.8%and87.2%respectively. In order todetect magolol and honokiol inMagnolia officinalissimultaneously, a method based on UVspectrophotometry wasestablished. But the results was slightly lower than actual content in herbs, so that thismethod would be used for preliminary detecting the content of magolol and honokiol inherbs.Prepared magolol and honokiol from80%ethanol extractin turn by ethyl acetateextraction,chromatography separationand recrystallization.Thentheir purity andstructures identidied by UV, HPLC and NMR, the purity of magolol and honokiolreached97.17%and92.93%respectively.The herb extracts, three mainly potentialantioxidant componentsand two monomeric compounds including water,60%and80%ethanol extracts, crude polysacharides, total alkaloids and lignans, as well as magololand honokiol, were the samles for antioxidant activity determination.2. Three chemiluminescence systems, respectively are pyrogallol-luminol system,hydrogen peroxide-luminol system and Fe2+-hydrogen peroxide-luminol system,wereused to evaluate the free radicalscavengingactivity of samples introducedabove,ascorbic acid was the positive control, and then,calculated the value ofIC50. Theresults showed that different samplesfromMagnolia officinalis had scavenging effectsonthree kinds of free radicals, especially thehydrogen peroxide (H2O2), which all thescavenging effects of sampleswere stronger than the positive control. In addition, theantioxidant activityof water,60%ethanol,80%ethanol extracts’on three kinds of freeradical was80%ethanol extract>60%ethanol extract>water extract, while therelationship between polysacharides, total alkaloids and lignans was totalalkaloids>lignansand>polysaccharides.In terms of the monomeric compoundshonokioland magolol,the former demonstrated much stronger free radical scaveningactivity than the other one, as well as the positive control. The results showed thatthebioactivity ingredients and compounds fromMagnolia officinalis had greatantioxidant activity so that this herb has a certain utilization and researchvalue.3. Designed and manufactured a microfulidic chip,whichintegrated eight parallelchannels and could maximum detect eight samples simultaneously, based onmicrofluidic technology and the theory of chemiluminescence reaction. Themicrofluidic chemiluminescence chip hadmany advantagesof simple structure, lesssample consumption, test fast and so on.Then, the scavenging effect ofmagololandhonokiol on hydrogen peroxide (H2O2) were determinateon the microfluidicchemiluminescence chip. The results showed that both magololand honokioldemonstrated scavenging effect on H2O2,consistented with the result ofmicroplatechemiluminescence detection system, which proved the feasibility of the microfluidicchemiluminescencechip system,as well as the potential application on the antioxidantactivity detection and screening.in the future. |
