Objectives:To observe the expression of CADM1,IL-6 and IL-8 in mice with liver depression asthma and the effect of Chaipu Tang intervention.Methods:thirty female Balb/c mice of clean weight?20±2g?were randomly divided into 5 group?A-E?and 6 mice in each group.Namely normal group?group A?,asthma group?group B?,liver depression asthma group?group C?,liver depression asthma+Chaipu Tang group?group D?,liver depression asthma+DXM group?group E?.Liver depression stage building:C,D,E group mice line 28 days random chronic stress?CUMS?,including the water,24 hours A day,24 hours of fasting,tweezers clip tail180s,bind bound for 1hour,swim 4?for 5minutes,thermal environment?45??,the level of volatility and reverse rotation,day and night for 2 hours and noise interference,living environment change?single cage,cage tilt,damp bedding,etc.?,A,B group of normal mice.Asthma modeling stage:group A was intraperitoneally injected with normal saline and atomized distilled water was used as the experimental control.Group D was injected with Chaipu Tang?1.5g/kg?0.3ml/only into the stomach half an hour before each atomization.Group E was injected with DXM?1mg/kg?0.3ml/only into the stomach half an hour before each atomization.Group A,B and C were injected with 0.3ml/only normal saline.Mice in groups B,C,D and E were intraperitoneally injected with a0.2ml mixture of OVA 10ug and Al?OH?3 400ug on day 1 and 8.On the21st day,each mouse was placed separately in a self-made atomization box,kept airtight,and stimulated by atomizaton inhalation with 1%OVA.Atomization was performed once a day for 7 days,lasting 30 minutes each time.After the final atomization for 24h,the mice were sacrificed with anesthesia and abdominal aorta bloodletting.The lung tissues of the mice were collected for HE staining image analysis.Expression of CADM1,IL-6 and IL-8 in mouse lung tissues was measured by WB.The relative expression levels of CADM1mRNA,IL-6mRNA and IL-8mRNA in lung tissues of mice were measured by real-time fluorescence quantitative PCR.Results:1.HE staining showed that the normal airway of mice i n group A had no special features,and there was no obvious infiltra tion of eosinophils and other inflammatory cells.In group B,the bron chial lumen was narrowed,the edges were thickened,and a few fract ures were observed.Inflammatory cells could be seen infiltrating aro und the lumen,and epithelial cells could be seen proliferating.In gro up C,the ling tissues of the mice showed obvious narrowing of the lumen,incomplete edges,obvious thickening,obvious proliferation and exfoliation of the lumen epithelial cells,and a large number of surr ounding epsinophil infiltration,No obvious hyperplasis was observed in the lumen of group D and group E,and the wall of the lumen was basically intact without obvious damage.Some inflammatory cell s were observed around the lumen.2.WB results showed that relative grey scale values of CADM1proteins in the asthma group and the liver depression asthma group were lower than those in the normal control group?P<0.05?,and the relative grey scale values of CADM1 proteins in the Chaipu Tang group and DXM group were higher than those in the liver depression asthma group?P<0.01?.The relative grey scale value of IL-6 protein in the asthma group and the liver depression asthma group was higher than than in the normal control group?P<0.01?,and the expression level of the liver depression asthma group was higher than that in the asthma group?P<0.05?.the relative grey scale value of IL-6 protein in Chaipu Tang group and DXM group was lower than that in the liver depression asthma group?P<0.01?.The relative grey scale value of IL-8 protein in the asthma group and the liver depression asthma group was higher than than in the normal control group?P<0.05?,and the relative grey scale value of IL-8 protein in the Chaipu Tang group and DXM group was lower than that in the liver depression asthma group?P<0.05?.There was no CADM1 was down-regulated and the expression of IL-6 and IL-8 were down-regulated in the liver depression asthma group.Chaipu tang can up-regulate CADM1,down-regulate IL-6 and IL-8reduce the infiltration of airway inflammatory factors in asthma,reduce the damage of airway epithelial cells,and relieve asthma symptoms.
