Objective:1To explore the effects of the Chaipu decoction on eosinophils apoptosisand siglec-8of asthma with Liver Depression of rat;2To explore the effects ofeosinophils apoptosis and siglec-8of humanMethod:1the effects of the Chaipu decoction on eosinophils apoptosis and siglec-8of asthma with Liver Depression of rat. Rat model of asthma with liver depression:70male SD rats, randomly divided into control group, normal treatment group, asthmagroup, asthma treatment group,the asthma with liver depression group, the asthmawith liver depression treatment group and dexamethasone group. The rats of theasthma with liver depression group, the asthma with liver depression treatment groupand dexamethasone group with chronic unpredictable mild stress(CUMS) in1to28days. The control group, normal treatment group, asthma group and asthma treatmentgroup received no treatment. Six groups of rats on the29thday of the experiment wereintraperitoneal injected with1ml antigen solution(10mg OVA,100mg Aluminumhydroxide gel). Strengthen sensitization on the36th day experiment. The controlgroup were given saline intraperitoneal injected. On the43th days,the six groupsexcept the control group were placed in the tank atomizer, atomized30minutes with1%OVA solution.The control group atomized30minutes with saline. Buxco animalrespiratory function measurement system to measure airway resistance in rats.Observed airway pathology used hematoxylin-eosinstaining.Observed the ratio of theairway inflammatory cells used bronchoavleolar lavage. Eosinophils apoptosisdetection was used Flow Cytometry. The human eosinophils was placed in10%HBS/RPMI1640, different concentrations(5%,10%,20%) containing medicine serum/RPMI16402h.Western-blot and PCR detection siglec-8expression levels.Results:1Establishment and Evaluation of model with Liver Depression: Thechanges of body weight of model with Liver Depression was significantly lower thanthe control group, the sucrose consumption was less than the control group, open fieldtest in vertical and horizontal was less than the control group, the still time of forcedswimming was more than the control group. The difference was statisticallysignificant (P<0.01).2Establishment and Evaluation of model with asthma: Compare with the controlgroup, the asthma group and the asthma with Liver Depression group was appearedrespiratory rate, rhythm arrhythmia, breathing like nodding, cyanosis, irritability anddull hair. This shows that successful of asthma model. In pulmonary function tests, theairway resistance in the asthma group and the asthma with Liver Depression groupwas higher than the control group(P<0.01). The hematoxylin-eosinstaining of lungtissue, the asthma group: many inflammatory cells infiltrated airway andsurrounding tissue, epithelial edema and shedding, bronchial mucosa and smoothmuscle proliferation, partially visible muscle damage fracture. The asthma with liverdepression treatment group: severe hyperplasia of airway mucosa and smooth muscle,visible muscle damage fracture, small airway obstruction, infiltration by inflammatorycell, alveolar expansion and accompanied by exudates. In bronchoalveolar lavage, thetotal number of inflammatory cells of the asthma group and the asthma with LiverDepression group wer(e78.00±6.88)×107/L、(89.5±3.69)×107/L, the data were higherthan the control group(33.00±2.58)×107/L. The proportion of eosinophils inbronchoalveolar lavage of the asthma group and the asthma with Liver Depressiongroup were(23.32±3.59)%、(30.79±4.62)%,the data were higher than the controlgroup(0.77±0.22)%(P<0.01),the total number of inflammatory cells and theproportion of eosinophils in the asthma with Liver Depression group were higher thanthe asthma group(P<0.01).In the lung function test, the airway resistance of theasthma group and the asthma with Liver Depression group were higher than thecontrol group, the asthma with Liver Depression group is higher than the asthma group(P<0.01).3the effects of the Chaipu decoction on eosinophils apoptosis and siglec-8of asthmawith Liver Depression of rat. The total number of inflammatory cells and theproportion of eosinophils in asthma treatment group were (43.25±3.77)×107/L、(5.07±1.29)%, the data were below than the asthma group(P<0.01). The totalnumber of inflammatory cells and the proportion of eosinophils in the asthma withLiver Depression treatment group and the dexamethasone group were (40.25±3.86)×107/L、(5.01±2.04)%,(41.50±4.65)×107/L、(5.65±2.59)%,these data wereall below than the asthma with liver depression group. There are no significantdifference in the asthma treatment group, the asthma with liver depression treatmentgroup and the dexamethasone group, but these data were all higher than the controlgroup(P<0.01). The eosinophil apoptosis rate of the control group, the normaltreatment group, the asthma group, the asthma treatment group,the asthma with liverdepression group, the asthma with liver depression treatment group anddexamethasone group were (5.59±0.61)%、(4.23±0.30)%、4.10±0.43)%、(5.74±0.46)%、(15.76±0.43)%、(17.18±0.66)%、(17.42±1.28)%,the eosinophilapoptosis rate of the asthma group and the asthma with liver depression group werebelow than the control group(P<0.05),the eosinophil apoptosis rate of the asthmagroup and the asthma with liver depression group were no significant differences. Theeosinophil apoptosis rate of the asthma treatment group is higher than the asthmagroup(P<0.01).The eosinophil apoptosis rate of the asthma with liver depressiontreatment group and the dexamethasone group were higher than the the asthma withliver depression group(P<0.01). There were no significant differences in the asthmatreatment group, the asthma with liver depression treatment group and thedexamethasone group. The siglec-8mRNA and protein expression levels of theasthma group and the asthma with liver depression group were below than the controlgroup(P<0.01). The siglec-8mRNA and protein expression levels of the asthmatreatment group is higher than the asthma group(P<0.01). The siglec-8mRNA andprotein expression levels of the asthma with liver depression treatment group and thedexamethasone group were higher than the asthma with liver depression group (P<0.01).4The effect of human eosinophils and siglec-8by the containing medicine serumof Chaipu Decoction. The siglec-8mRNA and protein expression levels of5%,10%,20%containing medicine serum of Chaipu Decoction group were all higherthan the control group(P<0.01).Conclusions:1The asthma with Liver Depression is mainly eosinophil infiltration, itsseverity is severer than normai severity.2Compare with the normai severity, theChaipu Decoction is more suitable for the asthma with Liver Depression.3Chaipudecoction can promote the expression of siglec-8and promote apoptosis of EOS inorder to control asthma with Liver depression inflammation effectively.
