Objectives:To investigate the correlation of the airway inflammation with the expression of MAL/MAPK/ERK pathway in the lung tissues of astimatic rats,to research the mechanism of chaipu Decoction in asthma disease treadment.Methods:Fourty SD rats were randomly divided into four groups,weight(220±20)g,each of ten rats,the asthma model group(group A),chaipu decoction group(group B),chaipu decoction and PD98059 group as inhibitor group(group C), and the control group(group D).Asthma rat model was establish by sensitization and stimulation with albumin(OVA)conjuncted with AL(OH)3. In group B,half an hour before to stimulate chaipu Decoction 2ml gavage(the concentration of 1.5g/kg). C group before challenged intraperitoneal dose PD98059 3mg/kg,Give the same chaipu decoction 2ml gavage.D group were given equal volume of saline spray and irrigation.The last challenge rats were killed at 24 hours after.obtaining lung tissue for HE staining and image analysis.Using PT-PCR to detect the expression mRNA of MAL,Eotaxin, IL-4,IL-10.Immunocytochemistry were respectively used to detect the level of p-ERK1/2,MAL,Eotaxin,IL-4,IL-10 in the rats' lung tissues.Results:Observation of rat lung tissue pathological section under the microscope.Rat lung tissue structural integrity and smooth lum en,occasionally(eosinophil)peripheral inflammatory cells infiltration,epi thelial cells had no obvious hyperplasia.there are a large number of eosinophils infiltration,part of the airway epithelium fell,bronchial s mooth muscle layer,airway wall thickening and mucus secretion.The symptoms of B,C group is less than A group.The MAL,IL-10 mRNA and protein expression levels in group D were higher than other group(P<0.05),MAL,IL-10 mRNA and protein in group A are in the minimum amount,The expression levels of MAL,IL-10 mRNA and protein in Group B,C(with Chaipu Decoction intervention) were significantly higher than those in Group E(P<0.05), significant differences were found between MAL,IL-10 levels in Group B,C and Group A(P<0.05). No significant differences were found between MAL levels in Group B and Group C(P>0.05), The IL-10 mRNA and protein were highly expressed in Group B were significantly higher than those in Group C(P<0.05).Conversely, the expression levels of the Eotaxin,IL-4mRNA mRNA and protein in GroupD are in the minimum amount, compared with other groups, there are very significant differences(P<0.05). The expression levels of Eotaxin,IL-4mRNA and protein was increasede in group A. The expression levels of Eotaxin,IL-4 mRNA and protein in Group B,C(with Chaipu Decoction intervention) were significantly lower than in GroupA(P<0.05). The Eotaxin,IL-4mRNA and protein were highly expressed in Group B were significantly higher than those in Group C(P<0.05).the expression levels of the p-ERK1/2 protein in GroupD are in the minimum amount, Tere was significant difference compared with A,B group(P<0.05),In group A the expression of p-ERK1/2 was increased, the expression of p-ERK1/2 was attenuate in group B,C after with Chaipu Decoction and inhiitor intervention. dignificant differences were found between p-ERK1/2 levels in Group A and Group B,C(P<0.05). The pERK1/2 protein were highly expressed in Group B were significantly higher than those in Group C(P<0.05).No significant differences were found between p-ERK1/2 levels in Group B and Group C(P>0.05).Conclusion:(1) MAL,p-ERK1/2 is involved in the development of asthma;(2)Chaipu decoction may regulate the expression of MAL, inhibit the ERK1/2 signaling pathway, upon which it inhibit airway inflammation in asthma.
