Object:Reasearch puipose:To explore the Chaipu decoction on asthma rats with Liver Depression,and the relationgship among mi R-21、PTEN and eosinophils apoptosis. Methods: Before model established, 70 Male rats were randomly divided into two groups: liver depression group(n=30),black group(n=40).Model group used chronic unpredictable mild stressing in 1 to 28 days,blank group have no intervention.After 28 days chronic unpredictable mild stressing, the weight were observed,sucrose consumption were observed,the blood adrenocorticotropic hormone measurements were detected evaluate the model success or not.After that, divide the blank group into blank control group(n=10),normal treatment group(n=10), asthma group(n=10), asthma treatment group(n=10),the model group was redivided into the asthma with liver depression group(n=10), the asthma with liver depression treatment group(n=10)and dexamethasone group(n=10).The asthma treatment group,the asthma with liver depression group, the asthma with liver depression treatment group and dexamethasone group,depending Ovalbumin repeatedly sensitizing and stimulating establish rat asthma model.Before the Ovalbumin sensitizing and stimulating 30 minutes,all groups rats received lavage treatment. The normal treatment group, asthma treatment group and the asthma with liver depression treatment group three groups received gavage in a dosage Chaipu decoction particles(Per gram particles contained 18.5g of crude drug)350 mg/kg;The dexamethasone group received gavage in a dosage dexamethason 1 mg/kg;The blank control group、asthma group and the asthma with liver depression group,three groups received gavage in a dosage of the same volume of sailine;After the last sensitizing and stimulating, all rats were measured the Bronchial asthma symptom score,animal respiratory function measurement system to measure airway resistance in rats,the pathological HE section of lung were observed, the the white cells,eosinophils and the ratio of eosinophils measurement in bronchoavleolar lavage. The bronchoavelar lavage Eosinophils apoptosis detection was used Flow Cytometry. Blood and lung tissue mi R-21 were measured by RT-PCR,Western blot, and each group the level of PTEN,m RNA expression were measured by QRT-PCR. Results: 1.Liver Depression identification :1)Weight comparison:The Liver Depression groups weighting 286.69±7.36( g) compared with normal groups 233.46±8.52(g) statistically reduced,with statistically significant(p<0.05).2)Sucrose consumption comparison: The Liver Depression groups sucrose consumption 8.41±0.79(ml)compared with normal groups 19.42±0.86(ml) statistically reduced,with statistically significant(p<0.05).3)Blood ACTH comparison: The Liver Depression groups the content ACTH in blood 1768±256(ng/L)compared with normal groups statistically 709±263(ng/L) is more,with statistically significant(p<0.05). 2.All groups rats of symptoms score and lung function results :The comparison of rats’ symptom scores and the pulmonary function tests airway resistance the asthma group and the asthma with Liver Depression group symptom was higher than the control group,with statistically significant(P < 0.05);The asthma with Liver Depression group more serious than the asthma groups,with statistically significant(P<0.05);The asthma treatment group was better than the asthma group,with statistically significant(P < 0.05);.The asthma with liver depression treatment group and dexamethasone group were better than asthma with liver depression group,with statistically significant(P<0.05);.There were no significant differences in the asthma treatment group, the asthma with liver depression treatment group and the dexamethasone group(P>0.05).The total white cells,the total eosinophils and the proportion of eosinophils in bronchoalveolar lavage of the asthma group and the asthma with Liver Depression group were higher than the control group,with statistically significant(P < 0.05);The asthma with Liver Depression group were higher than the asthma groups,with statistically significant(P<0.05);There is no obvious difference among the asthma treatment groups the asthma with liver depression treatment groups dexamethasone groups,without statistically significant(P>0.05).Lung tissue HE:Blank control group and normal treatment group lung tissue was normal integrity and smooth lumen airway mucosal edema, epithelial cells no obvious hyperplasia, occasional eosinophils, a complete normal alveolar septa. the asthma group: many inflammatory cells infiltrated airway and surrounding tissue, epithelial edema and shedding, bronchial mucosa and smooth muscle proliferation, partially visible muscle damage fracture. The asthma with liver depression lung tissue was more serious than the asthma group. The rest groups were significantly lighter than the asthma model group. 3.Flow cytometry instrument measuring each alveolar lavage eosinophils apoptosis in rats:Compare with the control groups(5.82±0.48)the asthma group(4.20±0.41) and asthma with liver depression(4.29±0.28) the eosinophils apoptosis rate of was lower than the control group,with statistically significant(P<0.05);The asthma group(4.20±0.41)the eosinophil apoptosis rate was no significant differences with the asthma with liver depression(4.29±0.28),without statistically significant(P>0.05); The asthma treatment group(15.74±0.34) was higher than the asthma group(4.20±0.41); The asthma with liver depression treatment group(15.77±0.42) and dexamethasone group(15.43±0.55)were higher than asthma with liver depression group(4.29±0.28),with statistically significant(P<0.05);.There were no significant differences in the asthma treatment group, the asthma with liver depression treatment group and the dexamethasone group,without statistically significant(P>0.05). 4.All groups blood and lung tissue mi R-21 relative expression The asthma group and asthma with liver depression group blood mi R-21 expresses less than blank control group and normal treatment group,with statistically significant(P < 0.05);The asthma group is more than the treatment asthma group,with statistically significant(P<0.05);The asthma with liver depression treatment group and dexamethasone group were expresses less than asthma with liver depression group,with statistically significant(P < 0.05);The asthma treatment group 、 liver depression treatment group and dexamethasone group three groups there were no significant differences,without statistically significant(P>0.05). 5.All groups lung tisses PTEN protein and PTENm RNA expresses The asthma group and asthma with liver depression group PTEN protein and PTENm RNA expresses less than blank control group and normal treatment group,with statistically significant(P<0.05);The asthma group is less than the treatment asthma group,with statistically significant(P<0.05);The asthma with liver depression treatment group and dexamethasone group were expresses more than asthma with liver depression group,with statistically significant(P < 0.05);The asthma treatment group、liver depression treatment group and dexamethasone group three groups there were no significant differences,without statistically significant(P>0.05). Conclusions:1.The liver depression model rats of asthma the blood mi R-21 expression is more than the asthma group, as well as the asthma symptoms and the degree of inflammation were positively association, mi R-21 may as the level of an important indicator of asthma control; 2. Chaipu decoction in order to control asthma with Liver depression inflammation,on the one hand can suppress the mi R-21 express,on the other hand promote the expression of PTEN,to induce the EOS apoptosis. Chaipu decoction may depend on suppress mi R-21-PTEN pathways inducting EOS apoptosis.
