Objective: This study screened the miRNA specifically expressed in the latent and active stages of Tibetan tuberculosis in the moderate areas of Qinghai,and verified the role of these differentially expressed miRNA in the incubation and activity of Tibetan tuberculosis moderate areas of Qinghai.Methods:Selection in September 2017-November 2018 who visits in university hospital,infectious hospital in qinghai province of Tibetan region(1500-2500 m altitude)height above sea level with active pulmonary tuberculosis and the 30 patients with latent outpatient,30 cases of healthy subjects,take a fasting blood in the morning and extract the RNA,then reverse transcription form cDNA,detection of PCR chip,to detect the expression of miRNA,Two specifically expressed miRNA(miR-181a-5p and miR-29a-3p)were randomly selected by simple random sampling method for qRTPCR,verification and functional analysis.Univariate analysis of variance(anova)was used for multi-group comparison.I use Fisher's exact test for functional analysis,use statistical software for graph analysis.Results:1.Twelve specifically expressed miRNAs were screened by microarray.Among them,6 miRNAs were specifically expressed in the Tibetan pulmonary tuberculosis group in Qinghai,and 3 of them were up-regulated(miR-151a-5p,miR-423-5p,miR-29-3p).3 down-regulations(let-7e-5p,miR-141-3p,miR-181a-5p);5 miRNA-specific expressions were observed in the Tibetan tuberculosis latency group in Qinghai,and 3 of them were up-regulated(miR-150-5p,miR-26a-5p,miR-25-3p),2 down-regulation(miR-142-3p,miR-181a-5p);2 groups of miRNA-specific expression in the Tibetan tuberculosis activity group in Qinghai,compared with the tuberculosis latent group,miR-425-5p up-regulated,miR-125a-5p down-regulated expression.2.Functional analysis was performed on the two selected miRNA.GO analysis showed that the predicted target genes were mainly enriched in protein synthesis,regulation of enzyme activity and regulation of transcription process.KENN pathway analysis showed that it may be involved in T cell signal transduction pathway,Wnt signal pathway,etc.Conclusion: 1.6 miRNA were specifically expressed in the Tibetan pulmonary tuberculosis group in Qinghai,and 3 of them were up-regulated(miR-151a-5p,miR-423-5p,miR-29-3p).3 down-regulations(let-7e-5p,miR-141-3p,miR-181a-5p);5 miRNA-specific expressions were observed in the Tibetan tuberculosis latency group in Qinghai,and 3 of them were up-regulated(miR-150-5p,miR-26a-5p,miR-25-3p),2 down-regulation(miR-142-3p,miR-181a-5p);2 groups of miRNA-specific expression in the Tibetan tuberculosis activity group in Qinghai,compared with the tuberculosis latent group,miR-425-5p up-regulated,miR-125a-5p down-regulated expression.2.Functional analysis was performed on the two selected miRNA.GO analysis showed that the predicted target genes were mainly enriched in protein synthesis,regulation of enzyme activity and regulation of transcription process.KENN pathway analysis showed that it may be involved in T cell signal transduction pathway,Wnt signal pathway,etc.
